laboratory grade autoclave

Quick question:
To sterilize my grain jars and pf tek jars I use a laboratory grade autoclave at school. But I'm having a lot of contams lately (bacterial) and I don't know how this could happen.
Now I was reading a fahtsters post and he said that letting your pc cool down to fast will result in your grain jars sucking in air. I'm using only polyfill as a contambarrier.
The autoclave runs for around 30 minutes and when finished the steam is drained. after maybe ten minutes the pressure inside the autoclave is the same as the atmospheric pressure.
My question is:
is the speedy drop in temperature in combination with only polyfill as a filter the reason for the contaminations in my grain jars. So far I haven't had any problems with my pf jars...

First off, make sure you don't get caught, cause that's a great way to raise some brows. A PC is a safer option all around.
Yes, rapid cooling can contam your jars if you took them out while hot.
The hot air in the jars when taken out cools, it's volume is reduced, and potentially contam rich air can enter your jars.
Always let them cool in the sterile vessel (be it a PC or an autoclave), that way the only air that can be pulled in is sterile air.
As for bac contams, that mostly dependent on sterile technique.

I for one do not trust poly. It doesn't sit right with me
SFD is the way to go 
One $1 SFD will make 10-12 dime sized pieces for your jar lids
and them shits will last you 10+ years and will continue performing to par consistently

I'm working in a high tech in vitro laboratory. I'm not trying to be smart here but why would 30 minutes be more than enough for every single substrate jar that is sterilized there. Why would 30 minutes be too short for sterilizing grain jars.

thanks for the concern but my colleagues know about my hobby, they just think it's for edibles. so no worries there

Then your answer is pc your stuff longer. Bacteria endospores are surviving the cycle.

Spores can take up to 2 weeks to germinate and start to show growth, then it can take up to a month for full colonization...this is a lot of time for contams to thrive.
Grains have endospores that must be killed...this takes time. Sterilize grain jars @ 15psi/250F for at least 90 minutes. 1/2 pint BRF cakes only need 60 minutes at this temperature/pressure.

I'm not quite sure if I'm allowed to adjust the cycles. If every cycle runs for about 30 minutes would it be cool just to run it three times?

there is not a single company that will ship sfd's to Belgium. believe me, I've researched it  I'm trying the dilated tek next with ironed polyfill.

normally after the cycle the steam drain is still closed. So when it says 'sterilized' I just open the drain (just a tiny tiny bit). but I think it would be possible just to run it 3 times in a row by resetting the autoclave without letting it cool down ...Quote:

haha almost did though. one of my first days in the lab I let it run without closing the steam drain. It ran dry for a while and started making weird noises. Luckily I didn't break it
