'My Growing Experience' - an entirely fictional short story in the first person by Higgle Pig.

06/05/14

Milestone: Began procedure.

Starting Implements: 2 syringes of B+, 1 of Puerto Rican
                    6 presterilized BRF jars
1 glovebag from a sponsored vendor
                    4 prepoured MEA agar plates
                    3 One lbs WBS bags
2 clear Karo presterilzed Liquid Culture jars
                    4 LC gauge syringes
                    1 Lysol spray can
                    1 Hot Pad (unneeded now)
                    1 hygrometer/thermometer
                    2 6 quart sterlite containers
                    2 16 quart sterlite containers

This approach may seem scattershot, but I am attempting to work on a number of fronts simultaneously. If I mess up one set of work, I will have at least two more possibilities that are not messed up. In keeping with this, I have retained one syringe, three agar plates, two jars, and have done nothing with any of my 1 lb WBS bags yet.

What I did do - prepare the glovebag. Used fresh, sterile needles for everything. First step: innoc one LC jar with some B+. This is where the first mistake occurred.

The presterelized LC jars had negative air pressure. By the time my glove encumbered hands realized that my syringe was getting sucked out, it had basically already happened. On one hand, this just means that that jar will colonize pretty rapidly (LC doesn't have the moisture balance issues that pumping a whole syringe into substrate would have). On the other hand, I wanted to use that damn B+ in the jars, not the Puerto Rican (which I got for free as a gift with my purchase).

Oh well. Live, learn. I innoc'd the second LC jar with the Puerto Rican, and used my thumb to prevent the suckage from taking all my spores. I used just a tiny bit on this, but the big black lump of millions of spores was the first to shoot through. I know spores are microscopic and that this isn't 'all' of them in the syringe (which I shook first) but it gives me confidence that 'plenty' got into the LC.

Okay. I don't have a magnetic stirrer. I'm starting out sort of small. These pre-sterilized LC jars don't have marbles or anything. I know you shouldn't slosh the stuff on the inside of the jar lid. So I have to settle for placing the jars flat on a desk, cupping my hand around it, and spinning it in little quarter turns back and forth rapidly like a washing machine agitator. But I'll get to this later.

Next, still in the glove box, I used the rest of the P. Rican to innoc one agar plate (I decided to save the other three for later - to test if my LC is contamed before hitting up my bags with it), then four presterelized jars. (Fresh, sterile needles). This glovebag was sprayed pretty liberally with propellant lysol in advance, and I'm dismayed to see the condensation of lysol is rather predominant inside the bag.

No matter, I finish my task. I put the foil lids (that were inside the seran wrap inside foil on my presteralized jars) back over the self-sealing innoc holes. I take a deep breath. And I unseal the bag.

I put the two jars in one cabinet. I put the petri dish down in a box with the innoced jars. And I wait.

Remaining Implements: 3 presterilized one lb bags of WBS
                      3 prepoured MEA agar plates
                      4 LC gauge syringes
                      Plenny of presterilized needles
                      1 syringe of B+
                      2 BRF jars

06/06/2014

Just about all I could hope to see today was the slightest potential for some kind of growth in the LC jars, particularly the one that swallowed my entire B+ syringe. I wasn't entirely disappointed on that front: small, white, faint bunches of thready material could be discerned in the jar. I did my quarter turn agitation and put it back. The P. Rican jar wasn't as thready at all, but I could see some whitish fuzzythreads on some of the big black spore clumps. Hopefully this is a good sign.

Nothing on the Agar yet. I didn't even bother to look at the BRF jars. Too soon.

I will continue to update this post with progress. I don't know about pictures, though. We'll see.

Alright, we're five days in. I was dismayed to find the temperature in the region was below what I intended. It wasn't shockingly low, just not ideal. Yesterday, I set it up with a hotpad under an upturned box to maintain the cabinet space at 79 degrees. This morning, I've seen HUGE improvement.

MUCH white mycelial strands are visible in both jars. The little blob of spores in the P. Rican jar are wreathed in clear white fuzz. I'm very pleased. Jars still show no growth as far as I can see. I didn't expect them too.

The agar plate, however, puzzles me. The lid is COVERED in condensation, on the inside. I am not sure what this portends. I worry about it. Could the lysol have possibly gotten inside somehow? Was the humidity within very high when I received it? I don't know how else it could have become so humid. Any input here would be helpful.

I plan to inoculate the wild bird seed bags by the end of the week at the earliest, but next week at the latest. I will be obtaining a few more WBS bags and a few more jars of liquid culture. I want to get several culture jars going while I have working stuff. I will just refrigerate what I'm not using presently.

Agar plate had no growth. Some kind of problem. Huge amounts of condensation. Chucked it as a lost cause. LC is doing GREAT. Going to inoc bags of WBS next Tuesday.

Three Jars are going great. Had to chuck one out due to contams. One is not developing but I'm going to give it another week before chucking it, as it is just inocc'd a couple days ago.

One agar plate innoc'd Friday is growing a splotch of white which is great! I fed the two LC jars flame sterilized air and used one to knock up six 1 pound rye bags. We'll see how that goes. Tested the less developing one on two agar plates. We'll see how that goes. So far, I'm pleased.

Worried that there is a lot of condensation in my agar plates due to the freaking amount of solution I innoced with. But we'll see. The white splotch in the first one is growing. How fast should agar plates grow?

Three of my four viable jars are doing great. The fourth is more recent and might be going great, but I shot my LC into the middle of the jar stupidly instead of the edge. I see one patch where something white is growing out. I will have to wait another day or two before seeing if that is a good splotch or a bad splotch.

Big progress!

After agitating the shit out of my LC jars, they are going strong with nice thick strands of mycelium. I will be hitting up some more grain bags with these in a week.

My agar plates have been removed from my heating chamber and the condensation cleared up after a day. A day after that, they are growing strong with mycelium! The only draw back is that the two that I used LC on instead of the first one, which was done a while ago with spores only, have growth all over the plate instead of one central location because I rolled the liquid back and forth trying to deal with the fact that I squirted kind of too much in there.

All four of my surviving jars are showing great growth.  The latest one was done with LC, which I foolishly injected into the middle of the jar instead of against the edges. But within a matter of days the mycelium has worked its way out to the edge in several sections and it fanning out in a nice, typical fashion where it meets the glass.

I have eight grain bags, three of WBS and five of rye. The WBS is done with spore solution and the rye is done with LC. Once again, I inserted the syringe stupidly into the middle of each bag instead of near the top so I can track grown. Unstupidly, three of the bags (a mix of WBS and Rye) are showing mycelial growth! So I have to imagine that the others will before too long. The LC grown bags are clearly getting a headstart - the jar done that way is going at a very accelerated rate compared to the bags.

Also, I realized I've been saying 'Peurto Rican' instead of 'Costa Rican,' which makes me look stupid. But in my defense, it was a free syringe and I just remembered 'Rican.' Haha okay, I'm stupid.

But this is going great so far! I'm going to get some more grain bags next week, a master grain jar, some more agar plates, and a scalpel so I can take the agar plates I have, grow some more, then innoc the grain jar with agar. Also, innoc some bags with LC. And maybe get some more jars. Remember: diverse approach for many paths to success!

Woohoo! All but one of my eight grain bags are showing strong growth. I gave up on the one that wasn't, and threw it out after sifting to visually verify that there was nothing wrong. Things are going great!

One of my four jars is almost completely grown over through the bottom, and is about to enter consolidation phase. It is pulling back from the jar edges near the top. The others are all close - even the one inoculated more than a week after the others with LC. (The earlier ones were done from MS solution).

My three agar plates are all growing at varying levels of decency. The best one, unfortunately, is showing a tiny spot of green on top of the mycelia. If that continues, I'm chucking it out (I opened the lid on that one when there was a shitton of condensation - a problem that was solved when the plates were taken out of the heated chamber.) I hope this doesn't spread to the other parts of the plate, but if it does, I'll chuck that plate.

The two LC jars are spreading still, shaking them up every day. Need to give them another GE burst with the flame sterilized air from the needle.

Going to move the grain to poo/coir/gypsum mix here in a bit, and use some perlite and foil to make a nice little FC for the four jars. That's not right away, but what's upcoming. Going to make a bunch of monotubs for the grain/poo mix.

I'm going to get some more grain to keep the process going. A master grain jar for the agar, and more plates. Thanks for reading about this!

In the meantime, I've got a bunch of 4-aco-dmt and a fuckload of HMRB and B. Caapi extract so I will have an easy time being patient hehehe

Alright, update.

Major developments.

One of my four jars was showing in vitro primordia a few days ago, so I birthed it, and dunked it in one of those 6 qt sterilite containers, with the other one on top and filled with books to weigh it down. Today, my vermiculite came and so I was able to roll that, introduce it into the 16 qt sterilite container with perlite on the bottom. I wrapped the lid in foil and set the rolled cake on that. It seems to be going great, and I'm misting it and fanning it regularly.

The next jar showed even more in vitro primordia, so I kept the process going and dunked that. The third jar is showing knots, but no primordia yet. It might be ready by tomorrow. The fourth jar is still colonizing (I innoced it more than a week later with LC, and it is almost caught up.)

So the jar phase is doing well.

The six rye berry bags are at about 70%. I broke up the mycelia and spread that stuff around and it's recovered and is growing well. I will be looking over their progress and after they finish colonization and consolidate, I'll move them to the monotub. I've got three 5 lb bricks of pasteurized hpoo mix (gypsum, etc). So that should go well - I have two LBS for each brick of poo.

The two WBS bags inocc'd with spore are doing a little slower. Some areas have great growth, but others do not. No where near as colonized. But it'll get there.

Got 10 one pound Rye Berry bags as well, and I innoced them with potentially too much LC each. I put up to 20cc each into each bag. That is very likely overkill. I'm hoping I merely have slow growth rather than having killed my bags with moisture. This happened just this afternoon, so obviously it will be a while before I can report about this.

Thanks, everyone, for reading. Anybody have any advice? Feel free to let me know what you think I could be doing better!

Birthed three cakes. Already had a harvest. One of those suckas was 7 inches long.

Did a monotub yesterday and a monotub today. Used a soldering iron for the holes. Worked well. I advocate that method. Used black garbage bags taped to the inside for liner. Mixed 2 lbs grain spawn to five pounds hpoo/coir/gypsum bulk sub. The one from yesterday is already starting to recover and spread. The one from today should do as well.

One jar is still consolidating. I'm waiting for some good primordia to pull that puppy out. The potentially oversaturated grain bags are doing good. Four of them were over 30% so I broke them up and redistributed it. Four are doing decently. And two are showing tiny white fans, but not doing as great as the others.

I haven't done anything with my agar plates but I might soon.

I anticipate having two more of my six initial grain bags ready to do another mono in the next two days. I need two more containers to do monos of everything I have now, but after that I just need to take a month or so off from hitting up grain bags and keep it to the tubs I have. I'm going to have more than I know what to do with as it is. No need to go completely effing overboard with it.

Yeah, I decided I can do pics. I'm sorry I haven't so far. It's just kind of freaky. But I'll post some tomorrow.

Got my last jar still consolidating. Going to birth it tomorrow and dunk the puppy no matter what.

My last birthed jar is pinning all over, including one really strange long pin that started as a big primordia, but it is drooping downwards. Very odd.

Harvested a ton of mushies from my cakes. I think I might just do a clean crop of them all tomorrow morning and redunk the cakes so they are all starting out fresh Friday morning at the same time.

My grain bags are all doing great. I did a 'test breakup' of my two-nearly ready for monotub bags and they are bouncing back nice. That's a last contam check before I spawn to bulk.

I am going to be getting 25 lbs more bulk sub here in the next week or two for when my big set of grain bags are ready.

I also ordered a food dehydrator to help with the drying of the harvests, but I have been running my small HEPA filter tower right on the mushies in a cupboard and they are desiccating really nice. After two days of that, I put them in a little tupperware container with a couple big packets of silca gel, but honestly they seem totally dry going in there.

I'm going to try out a batch this weekend, very excited!

And lastly, I got some Treasure Coast, more Puerto Rican, and some Malabar syringes. Put one each on an agar plate, hit up a master grain jar, and used a tiny chunk of a growing P. R. mushie to attempt a clone on the fourth agar plate.

Busy day! I have to do another Monotub friday.

It's been an awesome week.

I'm SO happy to say that the Monotub I did Monday is doing EXCELLENTLY! Little white fans spreading their lovely little wings all across the surface. The brown is giving way to the pale threads very nicely. Pictures tomorrow. The second tub seems to be recovering, but it was only 24 hours since I did it so I didn't expect to see much.

Many of those grain bags with the 20cc of solution are going VERY rapidly. I'll put pictures up tomorrow of all this stuff, like I said.

Thanks, everybody.
