Resterilization of substrate?
    #2012868 - 10/15/03 11:10 PM (17 years, 4 months ago)

Ok, my friend knocked up 24 jars using a homemade spore syringe. He also knocked up another 24 from a different syringe (homemade).. all on 10-12-03. Ok, now one batch of 24 is fine, colonizing rapidly.. but all 24 of the others aren't doing anything and have a slight odor... 90% sure it is contams. I have a stupid question now. In the faq it says that sterilization KILLS ALL bacteria. Since it has only been a few days, and since it is fairly safe to say that it was a bad syringe that is the culprit, would it be possible to just put them back in the PC and resterilize? Or should he go ahead and start mixing up more substrate? If it all needs to be thrown away.. should the jars be soaked in bleach water? if so, what ratio bleach:water?? thanks a ton

I guess you can argue both side. If you sterilize, all the contams will die, however they will still be left behind in the substrate. If this is good or bad I guess depends on what conams and how much. If there are no visible signs and just a slight odor I would resterilize and reinoculate. Especially if I was doing PF tek, thsoe jars are a bitch to prepare.

Yo,
What you probably have in your jars is a bacillus contamination. Look around some of the bigger grains of rice and see if they have a slight white line around them... thats a sure sign of bacillus. Not very visibily identifiable. I would suggest this:
Open one of the jars that has not grown and check for a smell of dirty ass/wet socks/supernasty. That also means bacillus. If it doesnt smell then just let the other jars set for a lil while longer. Bacillus spores are very heat resistant. So, I would not recommend sterilizing again at ALL, since now the jar is probably filled with thousands of spores. You might wanna try to do one jar, just to see if it works but its probably a waste to try to do all of them again. I would soak the jars in a 1:10 bleach:water solution for about a day and u should be good. Later,
~M

well.. other than the faint stench there is a SLIGHT discoloration at the innoculation spots on the side of the jar. The color is yellowish... looks almost like pee-pee.... smells funky... but there aren't many jars yoou can SEE it in... only faint smell as of now... think I should resterilize ASAP?

6C: you are right... these jars are a bitch...
