oyster?
    #1994275 - 10/09/03 06:59 PM (13 years, 3 months ago)

I pressure cooked a pint jar of mulch and sawdust then I inoc. with oyster spores. Turns out that there was a pool of water in the bottom of the jar maybe one fourth full. Is this too much water? Will this hurt the colonization of mycelia?

It could increase the contam chance but the oyster will still colonise. Id take the spawn from the jar and into other jars as soon as there is a big enough mass o' mycelium to work with.
Oh yeah and buy some agar.

Thank you. I have agar and have tried with no success. I always get contams and no mycellular colonization. I have tried making a glovebox and storing in the most sterile conditions possible but still contams. Maybe if I try a cutting on agar? I always seem to get a lot of condensation on the top inside of the petri dish. That might be a reason for contams but I have no idea how to stop it. Any ideas? After I spawn more jars I think I might make straw logs and fruit. I read somewhere that oyster mushrooms fruit very quickly when exposed to light.

You should go from birdseed ---> sawdust.
birdseed is much easier to colonize first.

thats it,
A trigger spray bottle with 20% bleach in it to clean air and surfaces in glove box before work.
Stack newly poured petri dishes to help with condensation then store with the agar surface facing. The agar will dry a bit if plates are in a dry environment so wrap some cling film round ther sides befor they get too dry. If you are getting contams round the edges try cling film from the start. H2O2 or a bacteriacide in the agar could help.
Everyone gets contams its just 'bout improving your ratio. Its normal to get contams when using spores on agar, you just have to run with it.
Yeah and oysters fruit as soon as you turn your back.
Dont give up on the agar and when you have it working do what bill said or go to the pasturised straw.... yum yum.
You are talkin like a botanist, but yeah a 'cutting' is a good idea.

Yea but how do you get that condensation off the inside top of the plate?
I ordered my agar from www.mycosupply.com

I've checked out mycosupply in the past.

250 Gram Bottle of Potato Dextrose Agar (sent to UK)

$24.95 + $57.60 P&P = $82.55

It costs me $9.89 to make the same amount myself

As far as condensation goes stacking your plates helps.

Storing them upside down also helps cut down on condensation.

Also let the media cool a bit more before pouring plates. The media container should be cool enough to handle without any discomfort before you pour plates, but of course don't wait too long!

Yea but how do you get that condensation off the inside top of the plate?

Stack the agar dishes when they are still hot and put a cup of hot water on top. The water vapor condensates only on colder surfaces.
See this url.
