karo colonization
    #1967638 - 09/30/03 11:02 PM (13 years, 3 months ago)

In others experiences how long has it took for their karo jars to colonize? I mixed 2 teaspoons of light corn syrup into 100 ml of hot water and stirred it around good, i steralized for 25 mins, let cool over night and innoculated with 1.5 ccs of EQ spores. Its been like 3 days since i innoculated it, does it sound like i did the procedure right? how much more time should i expect? Peace.

Yea, give it a few more days!! You should start to see some white hair looking things floating around. When left to sit it will all gather into sort of a ball. Give it a good shake to break it up.

Cool thanks, i got one more question. Its getting colder where I live and in my house its been low 70s the past few days, should i make an effort to keep my room like 75+ or will the karo jar colonize just fine at these temps.

Should be ok but I wouldn't let it get to cold or colonization might slow down!

I looked in the jar tonight, its about 3 or 4 days from innoculation, and i see tiny little things floating around that look white and fuzzy, little stringy stuff on them, this is colonization?

A FOAF has a karo jar going after he transfered a little bit of a PF cake to the karo/water solution. That was about a week ago, he checked it today, and found little islands of fuzz growing in there, but that's all. SO he gave it a good shake, and will check it in another week.

Question, even if the jar dosen't look full of fuzz, if someone was to shake the jar good, getting everything stired up, would the limited material inside be enough to start a BRF cake, or grain jar? We'er talking about a couple pinky nail sized islands here.

>> i see tiny little things floating around that look white and fuzzy, little stringy stuff on them

that's a good sign .
i've seen spores germinate in as little as 12 hrs. though 3-4 days is not unusual .
if it is at optimal incubation temp you should have a nice mass of mycelium within 5 days .

Cool cool, so one 100 ml jar of the solution with a good amount of mycelium in it should make 40 to 50 half pint jars right?

Ok cool, in the next few days i will have to prepare and steralize a lot of jars to be inocced because i just looked at the karo jar and there are many little clumps of mycelium floating around today, im happy =).

A FOAF waits until he sees about a quarter sized amount of Mycelium (doesn't have to be in one clump but rather all together) and when he is going to make some syringes out of it he fills the syringes full of Karo/mycelium and quickly squirts it back into the Karo jar then repeats one more time. This will break up the mycelium to microscopic pieces and that make every drop of that liquid culture a HOT shot!!!!!

Oh and if you can't use it all at once seal it back up with a sterilized lid put it in a Ziploc baggy and into the fridge with it. this will greatly slow the growth procees (but not kill the mycelium) therefore slowing down its consumption of the nutrients in the Karo and it will keep for months and months every one of my FOAF's Karo jars have been good to the last drop even 6 months after they went into the fridge!!!!
