What could be wrong with my karo jars?

I've tried twice now to create liquid culture jars.

Each time I prepared 5 jars with different sugars (honey, karo, powdered, granulated, light brown), 4% solution by weight with distilled water, PC'd for 30 minutes.

The first time, I birthed a BRF cake in my glovebox and transferred live mycelium to the jars with sterile (wrapped in foil and PC'd) instruments. No signs of contams, but the mycelium never grew out.

The second time, I inoculated the jars through a self-sealing hole (created with high-temp silicone) with spores from a reputable dealer. Again, no signs of contams, but nothing seems to be happening. Grain jars inoculated with the same spores at the same time are already showing growth.

Any ideas?

try pc'ing for less time,to much heat will caramelize the sugars,leaving them usless for mycelium growth.

If you're working with grain then why fuck around with liquid cultures? Colonize a grain master and do a buch of transfers. It's more efficient and less contaminant prone.

I also think there is too much pc-ing.
Try doing this one in the microwave.
150 ml of karo water in a tall half pint. Tight, but not drum tight plastic wrap over the top with 6 or 8 pinholes.
3 mins on high, 3mins off & repeat, remove & cover while hot with more wrap. Yes you will loose some as vapor.

Temperature could be a problem. Is your room too cool?

Or you could be adding too much h2o2 too soon or too little honey/karo.

Are you using tap water? Perhaps it is treated with something that kills fungus... I don't know.

If you are transfering myc instead of spores, add 20 cc of h202 to your honey water when you add your sample. This should kill everything but mycelium. Otherwise h2o2 will kill spores.

You could, even, soak the sample in h2o2 before you add it to the honey water.

If you shake your jars too much, you might not see growth. Give them a few days to sit before looking.

These jars need new air every couple of weeks. Give them h2o2 every time you gve them air. After two weeks they can suffocate (Unless refrigerated... cool air causes the mycelium to hybernate).

Most people have trouble growing the right thing in their jars. You can't grow anything. At least that means you're sterile.

Some people have indicated that they prefer liquid culture to grain-to-grain transfers. Anyway, I won't know which I prefer until I've done both.

Thanks for the extra info, folks. In none of the teks & posts I read do I remember anything about air to breathe. ;-) Doh. And it was suggested that sugars wouldn't carmelize in such a weak solution. Guess that was incorrect.

Back to the drawing board.
