sounds good to me. Unless you're isolating something that only likes to fruit in adverse conditions :lol:, I think it will work well though

Haha. True, that would suck if it only liked to fruit off agar! But I've heard it works well, so I'm gonna give it a try. Hopefully when I get it into a bulk sub it'll be a strong and plentiful fruiter too! I'm so excited to have an isolate, up until now it's all been ms which totally sucks!

the last two clones i took i purposely let fruits form in vitro in a brf cake jar.  i don't mess with cakes much, so i just did it up for a clean pin.  i just found an old creeper plate i should have tossed a couple weeks ago, maybe ill let that go.  i have a shiitaki pin, but culture is already an isolate

Just spawned a tub of cambodian clones that I took from a pin that popped up on a ms plate. Never had time to do much with it other than slant it up but, I will soon know if it was any good. Every plate that I inoculated with it sprouted pins in clusters so I got big hopes for it. Also spawned a few ms APE tubs that I actually started with. . . a ms syringe *gasp* hopefully they turn out ok, I have terrible luck with spore solution. Still trying to decide whether to G2G my purple mystic or just spawn the quart to a mini mono.

Gonna get another reishi grow going this weekend, turns out I got a lot of people in my life that can benefit from them.

Gnat problems are finally gone but its been a week with nothing fruiting which just feels wrong :frown:

yeah been busy with work and family, keeping the mycology going just don't have time to update much.  creeper grains are looking great and will probably do a g2g transfer those and some columbian rust spores this weekend.

Well simply that a pin that forms on agar from ms is usually pretty aggressive genetics. Most ms plates do not form pins IME. Every now and then you get lucky and hit a streak of ones that do but usually the plate just grows out and gets colonized. Pins that form without the plate being left at full colonization for some time are usually quite good genetics. Most genetics will fruit in good conditions but, the limited nutes, water, and FAE that a plate provides is usually not enough to bank on seeing pins.

IME I would say that less than 20% of ms plates will show pins, unless they are left at full colonization for some time. Usually I have forgotten about them by that point and moved on. If you do enough ms plates they will crop up enough to not be the most novel thing in the world, but I would not hold off doing a ms inoculation to get clones on the hope that the plate will pin first.

Frank!!! what do you think of my creeper dishes, I'm worried about the way they look. do they look like isolates? they're on the 6th and 7th transfers, not 8th like I had thought.

here's the latest transfers update. 10 days on the plate in ~65F.

I'm thinking that the tomentose growth is from being on the same media for 7 transfers. it's MEA for all of the plates so far.

I'm thinking the bottom middle and bottom left are isolates.

I have a few plates wrapped, in a box in the fridge  cuz i don't have the room to grow'em out right now.  I do have agar wedges from the second transfer growing out right now in two mono's though.  Once i get the space to use the next round, and get more plates poured, i'll drop a heads up here.

Your plates are looking great man, Cs57c looks great!

all four plates i had growing out from cultures which i thought were isolations weren't looking so anymore.  went back to the orginal plates, which were all taken from sectoring plates, and they still look like isolations to me.  so i don't know what's up.  regardless, i put agar to grain sunday and tonight they're fuzzing up.  testing three cultures, i'm growing three 3/4quart jars and three plastic cups, all brown rice.  stuck on this bulk vs cased grain thing, cause i want to master both of them.

fingers crossed, this grow will be my best yet.  thanks again bodhisatta.  and yeah, those plates are beauty.

FUCK!! i just re uploaded the wrong folder so now i have doubles of pics, and i am already low on space.  guess i have another thing to do tonight.

ok so here are three plate, all the 9th transfer.  all previous plates had three wedges per plate.  sectors sectors sectors no matter what i did, no matter how many transfers.  so to remedy that, i figured its the 9th plate, so i will do one wedge per plate, and let it grow out almost all the way, so i can take from the middle of a sector and get the iso.  i was taking really small wedges from the middle of sectors before this, but because there were 3 wedges per plate, there must have still been more than one sector each transfer.  so i will let these grow 2 more days or so, then transfer (this is KSSS from print)







also, got a little  surprise.  i had the worst worst bucket of contam, but it was a strain i really really have been wanting to grow (peurto rican) NEVER got it to fruit beyond pins, be it a mono, a tray, or a cake, or a v tek.  i have more jars, brf cakes AND v teks going now of it.  but the spores do not germinate on agar for me, so i have to either noc up a grain jar and transfer a grain to a plate, or take a fruit piece and put that to a plate.  trouble was the only bucket i got fruits from got super contaminated, it looked like this:



and whenever i took a pin, obviously there was bacteria on the plate.  so i tried a 10% bleach dunk, still crazy bacteria.  so i did an 80% bleach, 10% vinegar and 10% water mix, took 3 pins and SOAKED it for 5 minutes.  put three of them to a plate.  only one grew, but it looks like an iso or close to it: (those two black spots near edge of growth are the other 2 pins i dunked and did not grow)



needless to say, i will transfer that to one more plate to ensure cleanliness and then to a jar.  i have several prepped for different projects ready to be nocced up



also have a limited genetic KSSS bucket thats consolidating now, here is it with other stuff too (reishi and tazmanians in the other buckets)



and since it seems like ppl on this thread are sharing stuff in addition to what they have devoted for this thread, i guess i will jump on that band wagon

grain bag almost done to be spawned to mono



here is my new set up to avoid contamination.  three monos on left then shiitaki and enoki filter bags colonizing, an oyster bag fruiting and (i dunno if u can see) but three mini enoki jars spawned to fuel pellets in little v tek containers.  everything but the oysters will go into my GH as soon as i get it set up, hopefully tonight.



now that i see the pic in preview post, u cannot see the little enoki jars.  I'm still moving everything into that room, and have a couple more things left, just gotta get another table for it all, then i'll take another pic and show u guys.  i wanted some feedback as well.  guess ill set up the GH and then take a pic for better feedback
