Yeast contamination WBS vs Grass Seed * 1

So I had some WBS jars go down with yeast, but I also inoculated a couple grass seed pp5's on the same syringe.

But so far, almost fully colonized and no yeast white specks on the Grass Seed.

So question; does yeast contaminate grass seed like it does WBS?
or
Is it more likely that the yeast contamination was in the syringe, or that I improperly sterilized my grains?(I don't know how fully hydrated but not over-wet, pc 90 mins)

Would it be advised to throw a piece of colonized Grass Seed to Agar and see what happens? Would any underlying yeast be apparent on Agar?

Yeast very well could exist on the surface of either, it's how making one's own sourdough culture works (Kinda) (Admittedly typically one uses flour for that)

Yeast is killed by temperatures far below boiling, so it's not a sterilization problem.

Either you didn't follow proper sterile technique, or your overpriced pp5 containers didn't seal properly which is probably the true cause.  Use mason jars-they're cheaper, seal better and last far longer.  They're also made from sand rather than putting your cash into big oil.

I think you got my question completely backwards.

I guess I wasn't completely clear though so my bad.

The WBS is/was in Glass Quart jars. It got yeast.
The GS is in PP5, no obvious contams present.
Same Syringe.

*And the "overpriced" PP5's actually aren't overpriced.
24 "smalls" for $20.56 makes .86 cents a pop.
12 Glass Quarts goes for about $10 makes .83 cents each.
Competitively priced I'd say. Admitted I got a good deal on Amazon and didn't buy them in the grocery store. They might be closer to a $1 each there.

And to bring up Plastic and Big Oil... I think disposable petri dishes are a little more to worry about. At least these never get thrown away unless one melts which wont happen lol.
I think you just got something against anything Violet brings up lol.
BTW I like the PP5's cuz they're stackable and save a lot of space. Other than that I still use all my original glass jars as well.

Your WBS jars contam'd because you introduced yeast when you inoculated them.

Yeast will grow on all types of grains.

Your syringe is not the culprit since you had success with some.

So, after I flame sterilized the needle some yeast somehow made it through during incoluation, but not from the syringe...

So yeast was introduced how? I'm sorry but I'm missing something.

And grain is defined as wheat or any other cultivated cereal crop used as food, therefore Grass Seed is not a grain.

And since no other grains will be introduced in the spawning process with Coir and Verm, then any possible yeast wouldn't matter anyway.

Am I understanding correctly?

*Bottom line here, is the GS safe to spawn?

Injection ports are not fool proof. You can only wipe them with alcohol(sanitary not sterile) and you probably didn't inoculate in a SAB either.

Grass seed is a grain, you're just using agricultural definitions or wikipedia definitions, It's a grain to mycologists and it's a grain to farmers.

likely. Pollyfill sucks for injections, You can wipe it with alcohol all you want but you can push shit caught in the fibers right through. Injection ports suck IMO, all of them.

*Google Definition lol*Well fuck me. So yeast can hit the grass seed, and it didn't come from the syringe, but the glass quart jar with the polyfill got contaminated while the PP5 w/grass seed didn't, so some random fucking yeast fell through the polyfill on my WBS and my GS is safe if there are'nt any white specks?

I would in all likelihood assume yes to all of that.

I use polyfil for gas exchange exclusively on all my quart grain jars with no problem, I just don't try to inject through them. Yes you can use polyfil as an injection port just don't expect 99% of the time success.

ok well I transferred that supposedly safe GS.

I'm updating to tell you guys that the grass seed apparently carries the yeast with no obvious signs of contamination.
*Unless someone can direct me to a picture of yeast contaminated grass seed.

I transferred one pp5 of clean looking/smelling grass seed, which I had initially suspected to be infected due to an contaminated WBS jar inoculated with the same syringe. The WBS jar had white speckles everywhere while the Grass Seed had only healthy looking myc colonization.



Now I trnasferred the clean looking GS to 3 jars WBS and 4 pp5's GS.
All 4 GS pp5's LOOK clean.
All 3 WBS jars are contaminated with yeast.


Given to common connections here, I have to assume that yeast did not fall through the poly on my original yeast infected jar, and that my original assumption of a contaminated syringe now appears to be confirmed.

I did also throw a few grains to agar. From which I had two distinct contaminations on the first dish. One was some green, the other was small white splotches that didn't necessarily look like bacteria. I'm assuming that would be the yeast?

Lesson learned: Yeast can hide unnoticed in Grass Seed.

Now... Since I don't have much else going... to spawn or not to spawn.

I'm assuming that since mycelium colonization continues to occur, that there is still some fruiting potential. but possibly reduced due to the fact that the yeast has possibly used some of the nutrients itself, but with the myc colonizing over the yeast, maybe some of those lost nutrients can be reclaimed?

Interesting observation!

Spawning the yeast to bulk is most likely going to make your substrate hot, you probably won't get any fruits out of it :frown:
