Has anyone used UVC lights to reduce the risks of contams? I ran across this on the net and thought it would be interesting to share and find out if anyone had any experiences with it.

Never used them for sterilisation purposes but i'm doing some mutation experiments with cactus seedlings.

one thing: BE CAREFUL!

UVC is very harmful to unprotected skin and especially eyes. Leaving UVC lights on in an enclosed area leads to the buildup of ozone (explosive and toxic). Know what you are dealing with :smile:

Your mutation expirement sounds interesting Una. I wasn't thinking for sterizing substrate persay but rather the air in work areas say before innoculation or something.

substrate sterilisation won't work since UVC doesn't penetrate the substrate.

Yeah -- in work, all the time :wink:

It helps a lot, but unless it's only a couple feet from the surface it won't completely sterilize it by itself.  It really helps, though, in combination with other things (UVC lights are standard in the flow hoods, here.  That along with 70% Et-OH works great.)  The best thing you can do to prevent contamination is build on your sterile technique -- i.e. are you flaming the top of the agar jar between every couple of dishes?  You can also work in back of a flame (after cleaning the area) to prevent contamination from yourself or stuff outside of the bench.

Worse comes to worse, there are always antibiotics....

UVC lights in a flowhood, behind the hepa, can extend the life of the hepa by a lot.

seuss, I don't understand how they could extend the life. My thinking would be that the uvc would kill any airborne contaminants, but the dead bodies would still be in the air, getting trapped in the filter at the same rate as a hood without a light in the box.

I could see how the light would make a hepa more effective, first killing the contams before filtering them out, but that shouldn't make them last longer I don't think.

Most hoods I've seen with lights have them sterilizing the work surface, not the inside of the hood.

The UV-C kills any living microbs that get trapped within the filter immediately, rather than waiting for them to die on their own. In a wet or humid environment, the living microbs can actually eat the filter in some cases. I have a reference with pictures, lemme see if I can dig it up...

I use a 8W UVC bulb from an aquarium supply shop. UV-C is a very effective way of sterilising surfaces, transparent objects and clear liquids. It is not instant, there is some 'standing time'.

I use bulb with my terrarium ballast, no need to buy a 2nd, and put it inside a tin foil lined box. As has been said UV-C is very harmfull, it works by destroying DNA, so if you don't have a fondness for skin cancer you keep it covered :smile:

To work out the stand time, I am going to do the following experiment.

Fill a jar will a volume of pond water. Stand in the box taking it out very 10 mins and stiring (with a sterrile object), pour a bit onto an MEA agar plate, replace for another 10 mins, repeat.

Incubate the plats for a few weeks and look for growth.
