Breeding Clones
    #19504763 - 02/01/14 05:21 AM (10 years, 11 months ago)

Can you grow two fantastic shrooms from different MS innoculations, then clone and selectively breed them together on agar? Ive been doing a little reading on it, and you're pretty much just looking for a 3rd sector, correct?

I think you place two clones on Agar and wait for them to just contact on the agar.  If they are compatible they will fuse and mate where they touch and send off mycelium you want the aerial mycelium between the two strains and to select and clone those cells and see if it is a suitable cross.

Other methods I heard of were to time germination rates of the spores, add rattlesnake venom, or to heavily dilute and separate spores so they are intermixed enough during germination.

Another system all together is let many mycelial strains duke it out and get unique mushrooms and clone them.

Another system is just selective breeding.

Another system to generate mutants is to allow for certain competitors to take hold to force the fungi to alter its reproductive strategy and make selections there.

Another system is to grow on very compressed or odd substrates or cycle temperature wildly to get some unique clones to appear.

I haven't done much with any of this so I do not know.  That is where I'd start though.

You'd have to get 2 monokaryotic cultures and get them to mate on agar if you want to breed a strain. Workman also has a method of only isolating one monokaryotic culture, allowing that to colonize a jar then doing a MS injection.

I was under the impression monokaryon/haploid or primary mycelium would soon die if it didn't join with another hyphae and become dikaryotic
