First WBS jars looking good!
    #19501921 - 01/31/14 03:41 PM (10 years, 11 months ago)

This is my first attempt at anything other than cakes in a SGFC.  Knocked up two WBS jars with LC less than 72 hrs ago.

Thanks. I had them in room temp for the first 24 hrs, then built a TiT incubator and put them in there at 84 degrees a little less than 48 hrs ago.  I'm very excited!

Alot of people will say room temp is fine, but i prefer higher temps when colonising, they will love it in the incubator.

Its great fun doing your first wbs jars, much more interesting than brf cakes and the uses of colonised grain are endless! I love to experiment with things.

When you got LC, piss about, have some fun, make more LC, have more fun and most importantly......learn. If it fails, it wasn't for nothing, it taught you something

I can save you a bunch of time.

Chances of failure are the highest when making an LC from spores, grains or tissue.

Learn from other peoples mistakes.....skip LC and learn agar.

Ive had good success with LC and find it very simple.

I would like to venture into agar, but first, i would like to bulid myself a lamimar flow hood.

For now I'm just having fun. I have serious projects and then i mess about inbetween.

LC is sometimes frowned upon around here for it's inability to show contams unless tested, i love it however.  I knocked up some WBS with it, and with some agar brf sub wedges, and the LC is much further ahead.

I don't think you're wasting your time, I think any venture taken in this hobby will teach you something no one else can tell you.

EDIT:  I'd stay away from the 84 degrees man.  No higher than 80 is optimum:thumbup:

Don't let a $400+ investment stop you from dipping into agar.

I do not have a LFH, and have no plans on building one anytime soon.

A SAB and good sterile technique is all you need to be successful with agar.

I have done hundreds of agar transfers, and thousands of G2G transfers in my SAB and could not be happier with my results.

It is my opinion that new growers do not need to dump money on things like that so soon in their adventures....there are cheaper and better ways.

agar is the shit dude!  i'd actually say most ppl on the cultivation boards use an SAB instead of a flow hood, but i could be wrong.  building a flow hood just for cubes seems frivolous.  but to each their own!

I will definitely look into that, thanks.

With regards to the LFH i have all the materials here to make one, all i need is the filter. I have found a HEPA filter on ebay for £30 (20"x14"x1.5") aprox. So very cheap and...fun to make lol.

Get one that is at least 5-1/2 inches thick.

The thinner filters will not provide a laminar flow.

This is the most important part of the build.

Not to mention you have no idea what size blower you are going to need until you buy the filter.

The filter should be your very first purchase, then you match a blower to it.

I am also looking into the cultivation of King Oyster mushrooms on a large-ish scale, having a 8'x10' shed setup for fruiting 4' straw logs.

There is a good market around here for gourmet mushrooms and i have alot of local farm shops/restaurants interesred in a steady supply for the local posh people.

So a flow hood may come in handy in the long run.

true true!
i still have great success with my SAB.  actually have better (99% so far, only con tams most like from clone tissue, not transfers) but g2g less, like 85%, mainly because getting the last of the grain into the last receiving jars is a hassle, i always bump around a bit too much.  trying to find a larger tote for SAB, but none of them are clear
king oyster logs sound cool, only done blue tester logs, and small ones at that.

Finding them that thick is difficult, could several be sandwiched together? I've got a few spare extractor fans. The biggest being 10" also got carbon filters, ducting etc.

Ive experimented with small logs 12"x18" with success and cheaply too.

My local cheap/crap store has huge clear tubs 3'x2'x2' with lids for £15, i know where im going in the morning

No it's not.

Just search for 99.99% high capacity hepa filter on google.

In the UK, suppliers are a little thin on the ground. And cba to import.

A SAB may be the answer. Not sure if we have any air sterilisers here like lysol? I just mist isopropyl alcohol 70% in the air of my work area and wait a few mins?

Lysol does not sterilize, it sanitizes.

Chemicals sanitize, heat sterilizes.

70% iso is your best bet.

after i load my SAB with everything (that has been wiped down with 70% iso, paying CLOSE attention to the lip of the jars near the lid) i put lid on and latch one side, lift up other side maybe 6" and use spray bottle plain water, close lid, wait like 2 minutes, go to work!  Pusy farts design (i know it from pics he has posted previously) is a little better cause he has his upside down, no need for lid.  IIRC the floor of his is a shower towel , which is safer than mine, which is just paper towels on floor of tote, but the floor has grooves in it which the jars have a hard time staying still on at times when its packed tightly.  a shower towel i would think will grip them more, less movement when maneuvering jars during g2g.  he probably has a pic in his gallery.  just don't stare to long at his iguana....it blinks back after a while

A buddy and I have started nocing up mass amounts of millet for our grows with LC and some with agar. It seems to be colonizing fast and we're liking the idea of having so many inoculation points once we spawn to bulk. We have so many different things going and it's moving so fast we're going to have monos everywhere.

Most of the LC is stuff we made up from clean agar and put on stir plates for about a week and a half. A couple were edible LC's we picked a vendor so I suppose we are taking a chance on those. We also put them to agar. In all if we fail on a few its no problem. We have a really good source of millet right now and picked up 200 lbs at around $16 per 50 lb bag.

i make lc's with agar wedges in an SAB.  once grown enough, I suck into syringe and fill it, toss the remaining LC.  i test the syringe/lc on a plate, and if its clean, i store it in fridge, and flame before using.  kinda like edible lc's, or at least the one's i got.  i primarily rely on g2g, but sometimes ill use lc.

Sick setup, definitely going to make one of these.

How do you seal it with your arms inside? I can get vets gloves that go upto the shoulders.

Also, the lids of your jars, have you used silicone on top of the tyvek as inoculation points?

I have tyvek sleeves attached to the box, and I wear gloves.

The box does not seal completely.....there is no need for that.

My jars have no inoculation points....just SFDs.
