i hate waiting for spores on grain. maybe just my experience, but i have germination only 50% of the time (with syringe, so already hydrated), whereas agar and BRF i have success 100% with germination. spores to agar to grain FTW.
(sry if that what u guys weren't talking about)

Really? I've never had spores fail to germinate. We were talking about how slow galindoi is, so you weren't too far off

I have the same issue with spores to grain. A big part of why I use agar. Though I just cased my first mono that was started with ms solution to a single master. Thought what the hell and gave it a shot

I use to think "why not" when I thought about ms inoculation. Now that I've started with agar, it's a last resort and a risky one at that.

ok cool. well its strange, the only really problematic spores i've had were Peurto Rican (from vendor). never germinated on agar for me, only did on grain once outta 7 or 8 times, but if i used brf or a grain flour (other bob's red mill stuff) they always germinate. other spores (B+, Burma, Tazmanian, etc) only got like a 50% germination rate straight to grain, 100% onto agar. maybe I'm not using enough cc's or not shaking syringe well enough? or I'm just unlucky. just glad i got into agar pretty early. i was relying on a lot of g2g and lc's made with single colonized grain kernels to get my by before agar. after these next 4 tubs, and the grains i already have colonizing of cubes, I'm gonna post something in market place for possible trade for maybe pans or something of the like.

These aren't from spores. These are from agar wedges that have already been grown for a couple of plate passages.

Ok here you go. These are agar transfers from 1-16-14 on Rye Berries prepared according to RR tek. Because of the cold off the walls vs. the heated air there has been some condensation, but otherwise the grains started with the right hydration as far as I could tell. This is a pic of where the wedge ended up in one of the containers. There are maybe a couple other spots with a little growth.

This second one has many points of growth, but has nowhere near as much growth as other containers inoculated on the same day and even after using agar wedges of different cube varieties.

So let me know what you think. There is no sign of contamination that I can see so far. Just lots of grains hanging out with a tiny bit of growth.

well i woke up early this morning to check on my transfers i did yesterday which are all looking great! seriously a TiT has put my cultures into high gear!  BUT for the first time in a long time i found the mean green in my grains... seriously! WTF! it was only one jar so

all the ms transfers that weren't contaminated i held off until today and made some fresh PDYA, didn't have petri dishes so i just put them in ziploc containers, works great for stacking in my TiT though. so everything is now one transfer away from MS! thinking that might be as far as i go and make clones from these not sure maybe one more transfer and then to grains... we'll see how i'm feeling i guess!

The Pan tropics look like they're just starting to germinate nothing on grain The pan FL has no growth on the dishes or the grain The pan RDU has contam on the dishes just showing and on the grains The Reishi is kicking ass
The Creeper transfers I made a few days ago are going very slow and look faint, I can't even get a good picture of it but I suspect it's the 60F temperature in my house, there's some condensation that's formed on the dishes now too that I'm not very happy with either.
Here's two large creeper bricks unrelated to the creeper that's part of this thread.

Shit, are the rdu spores contaminated? Do you think they could be isolated away? I'm gonna put them on agar the next chance I get and I'll see if mine are too.

I'm sure the RDU can be isolated if it was warmer and the spores germinated already, I'll try again there's still some spore solution left for just that reason.
