Spore Germination on Agar questions

Hi all !

I've been using some drops of a spore syringe of mex (jalisco) on agar to experiment (I also did a diluted solution of 1/50 that I will talk about in another post).

I know everything is probably answered somewhere on shroomery but sometimes you want a custom answer and can't find all your infos :smile:

So on january 25 i made 4 dishes on MEYA or MEA i don't remember :smile:
The same day I used the same syringe for a few jars of rye.

1. I didn't open the petri dish and I can't see very clearly through the glass and little condensation, but what I see looks like solidified water like texture and not clear mycelium. I'm assuming there are millions of tiny pieces of mycelium and it does not give a proper appearance of mycelium yet, is that right ? It almost look like a weird bacterial contamination but again, I don't think it is :smile: Could someone descrbie how a few drops of spores look like after a few days on a petri dish ?

2. When comparing to the rye jars, I can now clearly see since yesterday that tiny mycelium is growing on the grain, but this isn't what I see in my agar :smile: Comments ?

I can attempt a picture but I don't want to open the petri dish and ruin it for the photo !

3. My goal of course is to try to isolate something interesting and i'll probably make another post about that part as I'm still confused about a lot of genetic notions even tho I keep reading paul stamet's chapter on genetics !

I think it may be too soon. MS syringe on agar can take weeks to get what ur looking for. I prefer to use a print on agar but sometimes you don't have a choice. Some tips I got is to use a plate that is dry to start off with and not condensated. Then when you apply the spore water, use only a well shaken drop and dig the needle in a very little in a short streak so the spores don't roll around on the top of the plate.

Hold on to those plates and wait a week and see what happens.

I get the feeling you are just starting in agar. It can be hard for people who've done it for a while to understand how overwhelming it can be when you're first starting out but trust me once you get it worked out you wonder why it seemed so daunting. All I can say is make lots of plates, be CLEAN, and take notes on the differences of each one. Even your agar recipe can make the difference. But it sounds like you're having fun and that's the important thing.

Thanks I will ! Just a question, why would it be better from a spore print ?

Aren't the spores better ready when they're hydrated in a syringe and isn't the risk of contaminant a little lower when you pour a drop of spores than when you drop flying spores with a loop ?

OH and can someone explain me why spore germinate and are more visible on grain than on agar ? I can see tiny fluffy things on the grain that are really not there yet on agar !

too much humidity / water with the drop content in the petri dish compared to a jar of grain ?

Yes to all your points. That is what is preached. However, IME, even old prints start easier than syringes on agar. It's like the water holds them away from the medium too long and they don't know they're not in the syringe anymore. I wish I had a better way to put it. But that's how I see it. Syringe plates always start responding once all the ambient moisture has disappeared and the spores could officially land on the agar.
With a print they get that signal right off and like you said the only road block is hydration but there is plenty of that to be had from the agar.

I am in no way an expert on this so don't take what I say as gospel truth. It's just what I've seen happen.

Everything that happens on agar is the same as with grain, but with grain you're not going to see the hyphae stage because it is a dark 3D substance where-as the 2D agar lets light through and makes everything very easy to see with the naked eye. Not to mention the agar is designed to barely be nutritious so growth is controlled and slower for observation.
