still-air box questions

Hi all,

I made myself a still airbox with gloves attached to it - it looks pretty much the same as - https://files.shroomery.org/files/13-23/032853058-18.jpg (from a tek in the 'general cultivation' section)

These are my questions, any advice appreciated!

1. For dealing with contaminants I used soapy water on the sides and bottom, but the bottom of the box isn't flat though so the water runs off and dries up fast. Does this matter?

2. If using a flame inside the box, can this be a problem because it disturbs the air?

3. There is a different type of box design where tyvek sleeves are mounted in place of gloves ( https://files.shroomery.org/files/10-47/097450939-GBOverhead_POV.jpg - from a post by TranscendingLife). What are your opinions on which design is better?

That is a glove box, and is not recommended for this hobby.

Build  SAB(Still Air Box), not a glove box.

A GB(Glove Box) is not the same thing as a SAB(Still Air Box).

A SAB is better for this hobby than a GB because the gloves are attached to you and not the box.

Making it possible to flame sterilize your tool outside the box between transfers.

Also when people try to "seal" their GBs, the spots that they missed(like around the lid) will constantly suck air in and out with your movements.

This will negate the "still air environment"....

1. remove the lid and use the SAB upside down on a wet towel.

This will give you a nice floor to work with, and the towel will create sort of a "gasket".

2. Never flame inside the box, always flame outside. Flaming inside the box creates heat currents.

3. I use tyvek sleeves in my SAB.....my gloved hands go right thru them and into the box.

Hi - thanks PF, that does seem like good advice

So you just use plain water then with the towel? (no soap)

Do you disinfect the air inside the box in some way?

I use a 5-10% bleach solution on my towel.

There is no need to disinfect the air, that is not the p;oint.

The point is to create a draft free, still air environment.

If the air is still and everything in the air has settled to the walls or bottom, nothing can possibly blow into your work.

This is what I used for pouring agar(from a small pot w/ lid) to plates and innoculating the plates from syringe. 10 plates I poured and innocd, not 1 contamd before they dried or till I started fucking with them (towards the later end of dish viability) I innocd 2 outside the SAB, full of contams within days bacterial growth n couple weeks, green mold formed. needless to say i got rid of those 2 quickly after that. Anyways I just washed it out with hot water on a cloth, i never contamed a dish in it and i dont use gloves and i really dont see why youd need it.

I just place two large paper towels on the bottom of the SAB, wetted down, and work off of that. It keeps the contams contained and the dishes don't want to slide on the plastic to liquid surface. When I'm done, I just put them in the compost pile. YMMV.

well it worked :O

anyways im just stating the way I do it, as is other people will do it another way. the media was sterile when it went in, and sterile when it came out. lol im not gonna try to convince you to not go full ass with it but half ass worked fine for me. all im sayin.

that was very similar to my first SAB you'll find yourself wanting to upgrade in a few months is my bet.:shrug:

I just put clean paper towels on a table, and put my box upside down on that.

gives a stable surface and more room.  much easier than trying to use the lid.

The purpose of the SAB it to protect the culture from you more than anything.

I give it at least 10 min. That gives me time to go over my procedure one more time so that I'm not making more moves than necessary. The less moves, the better as far as stirring up contaminates goes. That's just my personal experience.

But for real 99% of the suggestions here will work, except for the guy who doesnt glove up.  Ive never gotten HIV from fucking raw dog but my odds go up each time I do it.  Just like contams in your tubs/cakes.

Assuming the air remains still even when you're not using the box there shouldn't be many contaminants airborne inside it to begin with. I do know though that water impacting the surface can send some contaminants back into the air.

10m seems more like it, I've watched dust settle in a still room with a sun beam coming through the window, took a few minutes but it's cool to see when the sun is just right the air is still enough after you smoke or something through the curtains. you can actually get a sense of how the air settles it's really quite still after about 3-4 minutes then everything starts to slowly fall.
