You can... If youre going to be taking mycelium from the tub though i would recommend cloning your best fruit.

I would think that there would be a high risk for contams given that the mono was unlikely fruited in aseptic conditions... I'm sure that you could isolate a mono but it would likely involve more transfers away from contams to get a clean isolate. Better off taking from inner tissue or other way. My $0.02, curious to hear what others have to say...

well if im understanding this correctly

the mycellium/fruits are *1 strain* if i took a spore print from one of the fruits i would be playing the ms roulette again however if i took some mycellium/tissue from a fruit it would essentially pull the same traits as this same tub i have, correct?

I'm sure that even the mycelium taken from the mono would be heavily laced with spores (thereby m.s.) given that it is in/near the fruiting mushrooms... Mushrooms have evolved the ability to eject their spores remarkably well and to great distances; suffice it to say that spores have undoubtedly covered your entire fc (and contents) by the time even one fruitbody sporulates... To propagate on agar as you've described would effectively be an m.s....

Find a pin and put that to agar or find a fruit and put some of the insides of it to agar. Don't put the mycelium to agar unless you want to battle contams for fun and have lower vigor in your culture. One fruit from a MS grow is likely to have quite a lot of strains still in it. You could get lucky and get a monoculture/isolate from a clone job on a fruit but it's not that likely if you started from spores. If you clone it take one transfer then use that next dish if it's clean as inoculate.

no worries - wasn't trying to come off defensively... I don't find it hard to believe that some spores could have contaminated your fc pre-vail opening... I hope you experiment and post results - you should do both mycelium and fruitbody tissue (remember to rip open the stem then scrape a sample to avoid pushing contams into the inside of the stem) on agar and see which necessitates fewer transfers!  :whatdoyouthink: >>>(I'd still bet the house on higher success rate with a tissue sample or pin :rofl:)
