Spores "germinate" in pure H2O ?
    #18726857 - 08/19/13 10:13 AM (10 years, 6 months ago)

I had PCed one of the short jelly jars with a self heal port and double tyvek filter patch so it wouldn't get wet when tipped on its side. This jar was made so I could take a MS syringe and turn it into about 5 more syringes since I have found most vendors syringes are concentrated enough to expand reliably. This particular jar has about 20CC of sterilized h2o with some spores from the original MS syringe in it. Oddly enough inspection shows mycelium about the size of small snow flakes probably 5-10 of them floating around in the clear water. How come spores don't sporulate/germinate in the syringe but they did in distilled PCed water. The syringes I made from expanding it all were viable and produced no contams in the test jars I inoculated. It's like a LC without the syrup. I would attach a picture but there is no way my camera can resolve well enough to see these little snowflakes through the glass.

I have had spores germinate in a syringe. I have actually gotten them from a vendor with little white clumps. I had to shake the hell out of that bitch. I think storing in the fridge helps

Interesting, good to know. I usually use up all the spore solution I make but these jars were sitting with the colonizing jars for a week or so and it looks like a very clear LC now. I thought for sure I messed up something big time making those jars ha.

Well store in the fridge and I bet when you do go to use it that you will see growth very quickly

Spores will sometimes germinate using the nutrients in the water alone...I used distilled water for my syringes and store them at 40F to prevent this.
