Starting from spores on agar...? Pro's advice welcomed!

Hey everyone I prepared some petri dishes with some PDA agar. The bottle of premix agar I bought says to use 40g/l so for 10 plates I used 250 ml distilled water to 10g of the mix. I did 4 spore inoculations with a B+ print: (will get the other pictures later)


And then I took 5 clones, 4 of which I probably wont open because they look pretty bad, but you tell me =/
 
 

The only thing I think I would transfer off these clones is in the first picture, first clone. The print to agar looks pretty good and clean right now too, could I just put a wedge of that to grains once it grows out a little more? Do a few more tranfers?
I have nothing going right now And would really like to get some grains/tubs going again... I just needed something clean since my last tubs went bad to bacteria/trich...
All input is appreciated, and I'll keep the OP updated as my transfers take off. Planning on doing some work tomorrow after tonights feedback =)

I will usually do one or two transfers from the original print, just to be sure its clean. Its kinda a clusterfuck with the first plate because of all the different cultures growing.
Could probably get something usable from some of those clone plates, just make sure your opening them in a still air box so anything sporating isn't blowing all over.

Yeah that first clone picture looks decent, but I dunno about the rest, they look kinda ran by contamination... I'll do a few transfers today though and hopefully get some clean growth (:

Here's some of my transfers grown out, taken from the B+ spores. Some have nice looking sectors already, and others are lighter/more cottony. Probably take a transfer off of each, and further isolate...




I'd really like to put some of this clean growth to grains asap...Should I let them grow out a little more before taking transfers for grain masters? Should I just wait til I do another transfer and let that grow out before putting to grains?
Also should I be transferring from these by taking the very leading edges of the mycelium that looks more sectored or more cottony?

Take smaller samples next time you transfer. Grab mycelium from the leading edge of the growth and try to get a piece no larger than a grain of rice. This will cut down on the number of strains you get each time. RR
