All the replies seem spot on so I won't bother typing everyone.  replies over again. and yes I would make a brick or something out of the spinach container and then take the top off for fruiting and put it in a SGFC or similar.

I'll take a picture of the plates in a day or two. And get some reishi started

new variety of grains u mean?  i saw a good deal on a no corn WBS at home depot the other day.  it had black niger seeds, and i don't remember reading anything good or bad about them here on the forums...anyone have experience/input with that (black niger seeds) in their/others WBS grains?

bodhi-im excited for the reishi!  I'm sending u those pages within the hour.  please excuse the shoddy scan job..the book was thick and hard to press down on the seem.

Cool idea. You could either fill them full of bulk sub and spawn and then put it in a SGFC at full colonization, or you could tape two of them together to make it tall enough for fruits. I'd shoot for at least 4 inches of headroom. That should be fine if you've only got a couple inches of sub. If there's anymore, a few might bump the ceiling.

I want to see these grows. I supposedly got RDU in a syringe but Ijhave had zero growth on agar or grain.  Not sure if it just doesn't like my substrates or if it is a bum syringe.

thanks sarge and cronicr.  i like the bricks and trays ideas both, but can't fit many in the shotgun.  gotta make do.  i see a greenhouse in my somewhat distant future.

... but i did just figure out how to fasten one spinach tub upside down to another dubtub style.  i'm deffo trying this.

what is it about this mycology business that makes me keep finding new cats to skin?  such a fucking geek.  :rolleyes:  welp, got transfers to make.

It's official...It's The Mushrooms.  Didn't see that coming.    I don't want to misrepresent, it wasn't the mushrooms that  ended it, just may have been the last straw. 

My relationship with my mushroom closet, on the other hand, is rocking along smoothly.  I cloned out two each Shitakes, Pearl Oyster and Pink Oyster to agar.  Shitakes and pearl are grocery store clones, Pinks are from a live culture syringe (OutGrow).  All have gone nuts in the petri dish.  Both variety of oyster are almost to the edge and fluffy white.  The shitake are about half way to the edge, less fluffy but rhizomorphic nonetheless.

  I did a print swab and a dried piece of fruit body from a local mushroom (unknown).  The spores swab has shown no growth at all, good or bad, but the dried bit has thrown out mycelium in a rhizo pattern however there is a greenish mold standing on the fruit body itself.  I pulled it out of the closet and plan to go back into the glove box and transfer the clean mycelium to a fresh plate maybe tonight. 

  In the meantime, PC'ing away.  I've got a big load of WBS prepped and currently drying off, almost ready for the jars.

I'm happy to report that things are going along swimmingly.  Last night I made second transfer from the first MS plate.

It's bothering me however that these dishes don't have thick mycelium growth.  It's very thinly covering the surface of the agar with no visible rhyzomorphic growth.  It was mentioned that I might not be seeing that type of growth until a few transfers, so I'll chalk it up to that... it just seems out of the ordinary because I haven't seen anything similar through pictures, and it's a shame I can't share what I see with you guys since I'm using 1/2 pint jars. :frown:

I've inoculated 16 jars total, with only 2 strains showing thick cottony growth.  And this is with the same agar recipe, so I'm befuddled by the different results i'm getting with different spore syringes.  Anyone know what's going on here?  Maybe less spores in the syringes that are showing thick growth? :shrug:

Also, should I be doing more than one transfer from plates?  Or will I be doing that later on when I have less genetic competition on the agar?

Lastly, I'd like to say that it's awesome being a part of this, I'm enjoying reading up on everyones progress and happy that I'm not the only newbie participating. :laugh:

its normal for first plate to look pretty cottony during first stages of growth.  I'm sure if u let it grow out to nearly fill the 1/2 pint there would possibly be visible rhizo growth, but not necessarily.  there may be so many sectors that its impossible to tell.

i'd def take more than 1 transfer from each plate if u have the means to do so.  during those first plates when sectoring is hard to see, each wedge is a lottery ticket.  u don't know if its a winner or not, so the more tickets u buy (the more wedges you take) the better ur chances of hopefully getting a plate that has a visibly vigorous, fast growing sector.  I hope that made sense.  its also important because what if that one transfer/wedge you took got a contam thats difficult to transfer away from, now u r back to square one, unless u spot early/contam is small, then its not so bad.

To repond to your opening comment however, the mycelium only looks thick and cottony only with 2 out of 8 cube variety syringes. The other ones I can only describes as thin and translucent.  I was wondering whether the spore quantity per syringe would create a mess like that since you have thousands of spores each planting their flag on the nutritious media real estate. 

It should be noted that there was some condensation in the plates and that may have dispersed the spores all around the plate, I think that may have been the culprit for the results I'm getting.

Haha, oh man, I should just shut up and wait, just curious as to why this is happening. :p  I can only guess and throw it out there, hoping others have had this experience and maybe shed some light on the "why".

Looks like I'll have to clean out some plates so that I can get more wedges out from my MS Orissa India plate, I want that winning lottery ticket! :laugh:

yea u r right in thinking that!

the more spores u put down, the more germinate, the more they mate, creating dikaryons that branch out.  u want to limit that branch out, ideally in a perfect phillip k dick world, we could all get a print/syringe with 2 spores that mate and create an instant strain, but we don't.  so when we noc up a plate with a drop of syringe or a little scrape of print, or a swab of a q tip, who knows how many spores just got on that plate (i don't but I'm sure philip k dick RIP does or those who use Substance D :smile: ) ?  the less the better (as far as sectoring is concerned and our purposees here).  the fewer spores on the initial plate will mean less mating, less sectoring, less "jumbled up white cottony mess" (just on the initial plate if it is not grown out a long time, and sometimes even IF IT IS grown out to the perimeter of the plate).  perhaps, from the situation u describe, the 2 plates that DO display thicker growth coincidentally had fewer spore deposits on them...? 
as far as condensation dispersing the spores...not sure, never heard that or read about it so cannot comment on it (though i doubt it unless the "condensation" dropped onto the plate after it was nocced with spores and spread them around somehow whilst moving/handling the plate...but even then...would not affect the amount of spores on plate, just the amount of germinating spores reaching each other to mate/create dikaryons).  I'm no scientist though, so hope that helps a little.

Having a single strain formed from an MS inoculation (in the Dick world) would be so annoying...more often than not, strains formed during MS are not worth keeping.

As for your plates Whale, be patient. If you are not sure what you are looking at, take transfers to new plates. An MS inoculation always makes a "messy" plate, a transfer will show you what you've got in a few days time.

Condensation does not "spread" spores around, your agar media is sticky. Condensation on the lid does little harm, but you should try to minimize it. Keep your petris inside an insulated lunchbox while they are not in use. This protects it from temp swings, which cause condensation. Droplets that form and fall to the agar itself can carry contams from the lid as well.
