A few questions about GLC Procedures.

I use the Tek where you use a sponge and needle through the rvt silicon and have some questions.

When im going from jar to jar im not heating the needle at all. Im trying to keep the needle tip in the sponge but sometimes ill pull it all the way out of the sponge or push it to far before its in the silicon. Is that a issue at all? or as long as it only slips out for a few seconds before putting back into the sponge ok?im talking like a very small time of being out of the sponge like milli seconds.

also its Horrible when the needle gets bent at the end. Makes it hard to puch threw sponge and silicon.
Idk if anyone has a solution. I usually will heat it and bend it back the best i can.

Also what i have been doing with the syringes is when there brand new and in the wrapper there 100% sterile right? so i should be ok if i open the wrapper, throw on the needle and heat the needle and suck out some water i PC earlier out of a jar? sense the syringe is brand new i dont need to boil water and suck it up and such to get a clean syring right?

Also i have read that people loosen the lid a bit before the PC so that the pressure in the jar wont ruin the tyvek and silicon AE filter i put together. Is this true? I think last night i was a bit impatient and took the weight off the PC before it was even. I think i heard some silicon open and spew air from the jars because of the pressure difference it made like a squeeky noise. Guess ill see when they contaim lol. ( the tyvek square is on top of the jar. so i could imagine if the air is trying to go that way the silicon could give out on the sides. )

Is GLC Really worth the time and effort? Or should i just go to G2G?

These are a few things ive been running into.

I prefer G2G over GLC. Not that GLC can't work but i find G2G faster, easier and more consistent.

i would suggest you skip the sponge and just flame, yes i leave my jars loose during the pc and yes g2g is boss in my eyes

When you just flame arnt you worried about it being hot and melting the silicon? I loke the sponge cause in my mind it cools down the needle for the port.

you add another contamination vector with the sponge. Just squirt a drop of water just before you inoculate. I've gone in red hot before too without issues. The RTV just seals right back up

ok sounds good. So your saying that the sponge soaked in 91% alc is another contaim issue? i would think it would be better.... REALLY NEED TO JUST GET A FLOWHOOD!

70% iso is the better choice for this hobby but yes it only sanitizes and doesn't sterilize, heat sterilizes

no it's only sanitized not sterilized, it doesn't get any more sterile then red hot and you wanna use 70% alcohol not 91% for anything to do with this hobby:thumbup:

ok well at least i wont have to deal with the sponge anymore. Really thinking about going back to g2g tho... I just hate the GB.... its annoying. very annoying...... :frown:
