Why use "simple sugars" in LCs?
    #19408311 - 01/12/14 01:12 PM (10 years, 1 month ago)

Hello Folks,
I just secured a few grams of dextrose from a buddy of mine at school (they had a 100# drum of it!).

On the way home I got to thinking about something though.  Why do the LC preparations call for simple sugars like Karo, honey, or dextrose?  Why wouldn't you use a more complex starch, like maybe the starch you drain off from cooking grains?

The reason I ask is this.  First, fungi certainly can live on straight sugars (like fructose or dextrose) but bacteria readily eat them too.  If you used a complex carbohydrate, wouldn't that select AGAINST bacteria (most lack amylase and can't readily use it for energy) and give fungi a competative advantage in case you had some contamination?

The other question is, if I do a multi-spore LC, it seems to me that doing it into a nutrient solution that resembles the grain they will end up in would select for the spores that could utilize that particular nutrient profile.  I.e., you could make an LC broth by simmering a bit of brown rice and maybe a bit of quinoa, then straining off the grain and inoculating the (pressure cooked) starch solution.

I mean, people routinely use that water as an agar base.  I'm just curious why I've never heard of if used in LC!

Thanks for any thoughts.  I have dextrose so plan to use it, but I might try a grain extract LC just for shiggles.

90% of the time it's to make the LC as clear as possible. You can use any of the other things you said but they often get cloudy or dark etc.. Pretty much you never need an LC if you do agar anyway and you'll have more control so

the goal is to limit and control the amount of sugars we put in that will effect growth rates, the diffrence between 3 and 5% is night n day
