Hello everyone!

I am posting a thread to share some of the data I'm collecting as a third party for research on penguins. I hope that this little log can help out some first timers as my associate takes the plunge into the beginnings of mycology.


This is currently an ongoing experiment which we have not progressed far in quite yet, although the length of this document may hint otherwise.

Pictures will come when this place gets cleaned up a bit more.

Please keep in mind that my goal was to provide as complete of a report as possible; my job depends on it! As such, over time I hope to edit out any excessive or confusing information.

DISCLAIMER: There may be mistakes made with respect to the proper perspective of the scientist; I am only observing these trials and dictating their notes. I am not a certified mycologist, and any opinions I may hold are strictly from my own experience.


As of late in this arctic climate, the acquiring of cubensis mushrooms has been a rare event indeed, and even when you have an opportunity, they are often at unaffordable prices which cripple the penguin's wallet. My associate recalls a day when this was not the case, and has since decided to use their skills as a scientist to try and alter the current state of things, starting with this first trial run, and hopefully advancing to more complex aspects of mycology.

For the beginning grow, the decision was made [which, from my observations, is becoming quite common] to not only spawn to bulk initially, but to run two tubs concurrently. I realise this may not seem to be a good starting point, but there was no talking them out of it.


Materials:
PF Tek-
50x 1/2 pt Ziploc storage containers
5x 10mL spore syringes [Cambodia, Falbino, Panama, Peru, PES Amazonian]
1x bag of medium Vermiculite (size needed)
2x bags of Brown Rice Flour (size needed)
24 pack of bottled spring water
1x Glove box [60 qt., 23 1/2"x 18 3/8"x 11 3/4" w/ 2x 6 in. diameter arm holes, red lid]
-2x tyvex sleeves
-1x zippo [flame sterilization] (I need to get an alcohol lamp, I know)
-1x 750mL bottle of 70% isopropyl alcohol
2x large kitchen pots
24x canning jar lids
50 ft^2 heavy duty aluminium foil

Monotub-
2x Monotubs
-2x clear storage containers
[66qt., 16"x 26 1/2"x 12 1/4";
6x 1.5" holes for FAE (1 on top of shorter walls, 2 on each long wall, 5 1/2" from each edge) [See TL's Tub design]
1x bag of polyfill (size)
1x bag of coco coir (size)
1x bag of vermiculite (size)
12x 1 qt. jars [Bell, for pasteurization]

To be purchased:
1x digital thermometer
1x 5 gal. bucket
1x 6500k bulb

Procedure:
PF Tek-
1. Each container was prepared while watching RR's videos on repeat. There is a reason that everyone references his videos; every step was followed in earnest, and consequently this section will not be a rigorous review on how to prepare cakes. Watch the damn videos

25 cakes were prepared for the first batch [second is forthcoming], mixing 5 containers worth at a time. The recipe for 5 cakes was 2 cups vermiculite, 1 cup water, 1 cup brown rice flour, IN THAT ORDER.

Two extra jars were made as a control/timer; impatience is abundant here so to minimalise handling, these jars were allowed to be inspected and played with as an indicator of where the rest were at.

2. Large kitchen pots were used for steam sterilization due to the lack of a pressure cooker. As per RR a layer of jar lids, aluminum foil, then more jar lids were used to keep them off the bottom. There were three rounds of sterilization, water was kept at the proper level, and each was was allowed to sit for 100 minutes.

They were allowed to fully cool overnight, although I do not know if it is needed, and inoculation might be possible sooner. [If someone can explain when it is safe, that'd be  nice.]

3. The next day, 25x sterile cakes were inoculated, along with 2 jars, each receiving roughly 1mL of solution from the syringe. [The jars were inoculated with Panama & PES A, so those cakes got >1mL on average.]

4. Jars were allowed to colonise fully before spawning to bulk.

Data:
A. PF Tek
12/21/13 - 25 cakes & 2 jars prepared and sterilized
12/22/13 - inoculation; no major issues
1/2/14 - 2 cakes heavily contaminated with trich (disposed of), 1 slightly contaminated cake (isolated)
1/8/14 - cakes estimated at 60% colonization [i don't know anything and have awful spatial reasoning]

To do:
Prep & innoc other 25 cakes

B. Monotub
Tub #1 - 5x PES A, 10x Peru, 10x Panama
Tub #2 - 5x PES A, 10x Cambodian, 10x Falbino
(cubes are cubes, but i'm curious to investigate any difference in colonization time.)

Substrate (planned): Coir/verm/gypsum mix; 3" depth

Results:

Until the experiment is over, this will just be a collection of random notes and updates that will need to be taken into account when considering the experiment as a whole.
If enough plausible errors occur, it may be viable to start again from
the beginning while eliminating the errors.

-Some of the jars after being sterilized were left in the glove box to cool, which resulted in some signs of germination in Panama & Peru syringes.

It's hard to make a list of where every scrap of information comes from. I have done my best to keep a complete list of where all of my knowledge comes from, strictly due to my lack of experience and reliance on those who are willing to share their secrets. That being said, the closest and most effective replication of this experiment would involve a compilation of the following tutorials:

RR's PF Tek videos - basic cake preparation, sterilization and inoculation.
TranscendingLife's original monotub design.
Frank's list - substrate preparation, pasteurization, monotub modifications & fruiting conditions.

Well, thats one hell of a first post...

What are you using for a fruiting chamber for all them cakes??
or are you doing just monotubs??

Also, its not a great idea to mix a bunch of different cube strains together... I belive. Could be wrong on that one.

I would say do a SGFC to get some fruits, and use some of those jars for a couple monos.

I was planning to run two monotubs, I  purchased a second one which is much larger than I need it to be, but I figured that'd be good for the future.

To everyone else, y'all need to lighten up and laugh a little.
It's just a joke, I'm not going to feed these to penguins.


I believe tomorrow will begin the inoculation of the other half.
I worry about spawning to bulk with 3 lost to contams and a handful that are super stalled. I feel like it may have been the water content? Most have developed some condensation in the containers themselves.
Meh. who knows.

so, several of the cakes have reached full colonization, however a portion of them have some time left or have stalled.

at what point should i begin the spawning to bulk process? after some other cakes catch up?

Apologies for the communication blackout. Had some struggles due to penguin mating problems.

Brief update, the monotub was a sham, lost it to some sort of bacterial contaminant. There's a wide margin where procedural errors were made so next go the process will be a bit cleaner and faster.

The other half of the cakes were spawned to a SGFC.





It's so damn cold near the South Pole, it's crazy. Temperature has been an issue due to energy limits placed on the lab from the higher-ups.

better update needed. too high for all that right now.
