Understanding Cloning, Isolating and Mycelium Behavior

I have a general idea of the processes and functions of cloning and isolating but was curious to hear how others go about it all. I still have questions and am curious about how the mycelium behaves and just general understanding of it all.

This is what I have gatherd. I know there are many ways of doing things and would like to hear them and the reasons behind them.:cool:


OK
Lets say your starting with...




-MS jar spawned to coir/verm

- Select an early growing, good looking Pin from a cluster perhaps
OR

Clone tissue from inside of the lower part of the stem. 

Transfer to agar (MEA). 



----  Now some say its better to isolate if you have sectoring while others say the "combination" of sub strains should be kept :hi5: "together" as like one synergistic organism that makes up the fruitbody, I guess.. This is interesting but confusing.

How would you keep them together, just transfer an edge as soon as you can instead of letting it grow out to see sectors?

-Anyway, so you make another quick transfer to agar and let this grow out. How far? an 1" in all directions?

-Select a leading edge and cut a little piece out for grain jar or (if its a keeper) a master slant. 

Would it be easier to just refrigerate the petri until you know if its even worth saving and if so transfer into a master slant?




-So the clone is grown out and fruits.  If you didn't isolate the sectors out your going to have a few sub strains still but are they going to be "fused" together and acting like one. 
OR
 
Are you still getting different sub strains on each flush just like MS but maybe less and more cooperative with each other? Is that about how it goes...?




-OK so you like what you see and want to "save" and perpetuate the clone.  Are you going to take more clones of the clones now or at any point
OR

Just use the original agar dish in the fridge to make a slant and dip into that whenever you need it?

the sectoring thing is up to you how you decide to go about it but if you do decide to transfer sectors i transfer all of them and fruit each but i haven't really run into any crazy sectoring, a little uneven growth for the most part but i don't even pay much attention to it(i probably should lol) i just transfer away from the tissue and my growth has always been great right after anyway:shrug:

and i don't ever clone from a clone i go back to the dish, i'm trying to hunt phenotypes right now so i will be cloning and paying much closer attention/not isolating sectors-fruiting and starting from prints and looking for those genetic traits again and cloning and repeating but thats adiffrent story all together and a different q:wink:

and after i make my first transfer from the clone i let it grow out as much as possible as i'm not taking anymore transfers
