i don't usually fuck with lc now that i have agar, but I've been bored, so i make tall 1/2 pints lc's for one time use (nocced up with a clean agar wedge), then toss. qt jars r too big to use for lc's for me. actually, taking an idea from cron's agar dishes, i think my next lc will be a mini one made from a spice jar, so its even smaller. the closer u can get the needle to the myc in the water without really messing with the jar, the better, IME and a mini spice container is PERFECT for that.

These pp5's are great man...small, stackable, and you can see right through them....

The one on the right is a glass jar, but it's pretty difficult to make out what's going on in there.

so just used the lc i was talking about above, first time making one with no GE hole. PC'd syringe wrapped in foil 20 min with air inside (20ml) planning on using 10ml lc 2ml per jar (5 jars if u do ur math right). DAMN that shit had a vacuum like no other, even with the air in needle. i thought it would suck the air out of the needle as soon as it penetrated the RTV, but it didn't. i tried sucking up the lc into the syringe without expelling any air (it was sterile after all) and nothing would suck up, but the plunger WAS able to be pulled back. it was not until i expelled the air in syringe into the air of the lc jar that i would suck up any liquid, and even then, i could only suck up 7 ml of solution. weird. I'm no physics major though (though i can fly like a pig!). maybe there was a breach in the lid or rtv. also put a drop onto some agar, why not? only had one PP5 left to use, and it was between testing the lc (despite already knocking up jars) or taking clone flesh from a non sporulating fruit. maybe i made the wrong choice

The way I understand it, if you have a mason jar with a rubber seal, when the air heats up it pushes the lid up a little as the air expands, letting air out. Then as it cools it pulls the lid down and the air continues to shrink as it cools, making a partial vacuum. So you have to add a bit of air, I guess more than 20ccs in your case.

You could avoid all that if you drill a very small hole in the lid (besides the SHIP hole) and cover it with some tyvek/SFD/or E felt. You could cut the tyvek same size as the metal insert of the lid, remove it to shake it up, so it doesn't get wet, and replace it when you're done so it can sit on a shelf protected.....

You never remove your filter, and if you're gonna use a filter like that, you have to have a stir plate or else you'll just get a glob of myc that won't go into a syringe.
Use a Whatman filter, or use 2 injection ports and an airport syringe.

You can cover the hole in the lid and shake the LC jar to break up the mycelium. The only time you have to remove the filter is when you do this, otherwise it will get wet, and that's a no-no. So you don't really need a stir plate, or anything fancy. You can also safely remove the filter to facilitate this, and successfully. I've done it, and i'm a noob, so if i can, he/she can.
I placed a LC jar in SAB with nothing more than a tin foil lid on it, covered the hole with tape, shook the shit out of it, and used a sterile syringe to draw up the culture. I then used that culture to knock up 8 BRF agar sub jars, and all 8 are growing....they're still babies, but they're growing. No SHIP, no filter, no nothing on the LC jar, and it worked. So rather than getting into injecting air in a jar to stabilize pressure, it would make more sense to use a very tiny hole to do that for you, and cover it with some sort of filter for when it sits there doing it's thing.
Go to page 7 on this thread and watch me do it.

I took even less precautions than mentioned above, though i will def use a SFD with a SHIP next time i do LC

Again, you NEVER remove your filter. Just because you got lucky on 8 bags doesn't mean it's a reliable method. This kind of thinking will ruin your future grows.
Do 800 and then come back and tell me how it goes.

THey weren't used on bags man..i used the LC to knock up small pp5 jars. IF you say not to remove it, I'll listen to you. You seem to know what you're talkin' about, based on some other posts i've read of yours. I'm glad I got lucky on those 8...cheers
