Chaga fruiting on the plate!
    #19366581 - 01/04/14 01:10 AM (10 years, 1 month ago)

Crazy as hell!  This chaga was cloned from fresh sclerotia tissue and has now made fertile polypores in vitro. Dropping spores like mad. Dunno what to do next exactly. Agar is mypa. Might make a bunch of multispore plates from one of these.

Very cool AV. At what temperature were the fruiting bodies produced?

The lab stays at about 65 F.  These are at five weeks from original clone. Never subbed and somehow completely free of bacteria.

What should I do y'all?  Sub out little pieces of the fruit?  Or multispore?  My goal is to get vigorous Myc for plug innoc but I do not fully understand the life cycle.

Looks like you've got an amazingly vigorous culture for your plugs - can't you just transfer parts of it that aren't fruits and use that to colonize your plugs?  Sorry maybe I'm confused about what you're asking.  That's an awesome culture though, mine sucks and grows super slow and doesn't do ANYTHING like that.  So I wouldn't see any reason to go to spore, really...?

I was reading the other day chaga will often produce fruit bodies and spores when it's tree dies, as it's last chance to pass on genetics, so maybe your dish there felt threatened

Maybe try to take a few samples of the mycelium @ 5 o'clock in the picture above to get as far away from the spores as possible.  Then get a few more samples at random areas and compare P2 plates, all done in a SAB, of course.  It would be a shame to loose those original genetics!

Duh, the spores didn't even occur to me   Good point.

Yea. The spores pretty much contaminate the original culture strain. This may not be a bad thing though. I with I would have subbed it prior.

I would imagine a transfer of the culture (one as fast as yours) would outrun the spores before they could germinate and contaminate it, wouldn't you think?

 That sounds like it's worth a shot, Av, maybe try a non-nutritive agar to make the Mycelium run even faster, and make the agar a little on the dry/firm side by adding a little extra agar agar powder.

*/* P.S.:  When you have what you think is clean mycelium, isolate some of it to sterile distilled water for long term storage.

Good idea y'all. Will sub away tomorrow. Don't wanna get too carried away on this project. But it was cool to see the fruiting bodies for sure. I had never seem them in nature.
