precipitation on top of petris
    #19363170 - 01/03/14 12:21 PM (10 years, 28 days ago)

i am uskng both baby food jars, the smallest type, labels removed, sterilized in pressure cooker at 15 PSI for 45 minutes. All cultures are in a well sealed inoculation chamber.  I believe much of the contamination is due to a precipitation that forms on the lids of the culture dishes. So far the only nutrient media that doesnt form precipitation, due to it cooling quicker than PDYA or MEA, is Oatmeal agar, but I havent seen any spore germination just yet, after five days.  What can I do to eliminate this precipitation.  Do I have to put a little water in the petri dishes when autoclaving, or can I just set them in there?

You can use a filter gas exchange port in the lid. That would help reduce
the amount of condensation as the water would have somewhere else to go.

When pouring plates you want to wait for the agar mix to cool quite a bit before pouring. That will help. But from what I can tell you are putting in
the agar mix into the jars and then PCing them.

After you inoculate a jar with spores store them upside down so the condensation is not on the agar.

If you stack the petris when you're not working with them you'll get less condensation. I imagine the same is true when using baby food jars.

The condensation forms in empty petris i put in the pressure cookers, but if i stack them inside PC and wait for them to cool, there will be less precipitation?

Only when there's something growing in it. A little condensation during the sterilization process is pretty much unavoidable but shouldn't cause any problems. If you let it cool enough the condensation will have evaporated.
