anyone run into this ubiquitous scumbag?

This contaminant was growing in PDYA, it doesn't seem to form on Oatmeal Agar.This is from a spore jnnoculation, not a plate to plate transfer, and the syringe was flame sterilized and cooled in a blank agar tray before innoculating. The petris are placed in an inoculation chamber with arm holes. I wear gloves, a respirator from an old job site i worked, and a full body tyvek suit. I haven't seen pictures of this anywhere i've looked. I'm beginning to wonder if it is a defect in the agar media itself, maybe a problem of proportions or of substandard potatoes....anyone, please, can you help this noob.

Yeast infection. Right side of the petri is probably bacteria.

Thanks, something in the back of my mind kept telljng me that. Standard methods of sterilization will eljminate, or is it difficult to get rid of? Is it dangerous? Most importantly, it seems to inhibit mycelium growth, so prolly throw anything showing signs of this away, huh? Could it be that I am adding yeast from packages that were left open?

It's annoying, but it's not dangerous. It's everywhere, spreads fast, and inhibits growth of other organisms. Standard sterile procedure should be enough, but now you know to be extra careful.

That's all bacteria. It's the kind of growth I would expect from some Bacillus species. I think you'd have a lot better luck inoculating the agar with dry spores from a print instead of the syringe.

Well it's hard to say but it does look exactly like a Bacillus subtilis colony which coincidentally is the bacteria you'd most likely see if the agar hadn't been completely sterilized.

You are very likely right! I've been comparing some pictures and most yeasts seem to wrinkle differently. But some of them look very similar to B. subtilis!

If you still have some of the PDYA left I would resterilize in the PC before using it again just in case. Otherwise normally after agar has been sterilized once you'd only need to heat it enough to turn it into a liquid so you can pour it.

Thanx for the info. I removed all contaminants and bleached all surfaces and changed out all cultures, leaving only those utilizing Oatmeal Agar, and this is the third day and have noticed no Bacillus growing yet. Think I'll sterilize the lab every day now, though.

Before I thoroughly resterilized the lab, i had inoculated plates from the same spore syringe using both PDYA and Oatmeal Agar. The mycelium loves oatmeal, it seems like it finally took of rhizmorphically on this medium, and not a single one became contaminated with Bacillus. I believe the Bacillus gained entry when I started running spawn on grain. I think it is a thermophile and so I should have soaked the grain prior to autoclaving. It would be curious to confirm if anyone else has had the same experience with oatmeal agar, in terms of a possible preference Bacillus may have for PDYA. Anyhow, thanks for the help in identifying this.
