Basic syringe sterility concern (liquid culture)
    #19358515 - 01/02/14 01:21 PM (10 years, 28 days ago)

Lets say you have a syringe and you pull it out of the PC and your ready to suck up some lc.  Now image the part of the syringe shaft, the inside of the shaft, the part that is exposed to air now because the plunger is fully depressed.  This part is exposed to any contaminant that may be in the air.  As you take up LC the rubber gasket is sliding along the inside of the shaft, so how do I know endospores aren't slipping past that gasket and contaminating the lc?

It seems like they would.  I thought about stuffing some pillow stuffing in those gaps before pcing the syringe for extra cleanliness.
Thoughts, suggestions?

never seen someone do that but i guess it could work.  unless its an old syringe, i wouldn't worry bout the rubber part letting anything thru.  i just keep it wrapped in foil till ur rdy to flame it.  i have 6 month old edible lc syringes still in fridge in original syringe that work fine, and they can't be PC'd

You would flame sterilize the needle right before you penetrate the SHIP......

If you are that worried about it, you might want to forget about LCs and focus more on agar.

I will never use a needle again.......

No contamination of 120 samples was found after the ribbed plunger shaft was grasped by a bare, unprepared dry hand during five cycles of filling and injecting the medium into the vials without removing the needles from the stoppers.
