How long do you let your needle cool for after flame sterilizing?

I'm curious because on the one hand I don't want to scorch the spores especially if I'm just doing one drop for agar work.  On the other hand the longer I wait the more likely it is that contams will land on it.  I suppose the only way to really know is to take a laser thermometer and point it at the needle.  Has anyone done this?

I apply red rtv to the 'bottom side' (the part inside the jar) of red gaskets and use those as my inoculation ports. When the rtv has cured after application (up to 24 hours) you can inoculate. 

I flame sterilize, and while still red hot, I press the needle through the port and then let it cool by squirting liquid through the needle. Easy peasie. 

After some time you will need to apply new rtv but it takes a while if you did it correctly the first time you applied it.

I agree I squirt a few drops then take the last drip left and touch the agar. If you have problems with that then cool in the receiving agar put a drop. Sterilize repeat. Try them all pick what works best for you.

Well most people use alcohol to wipe a ship before inoculation.  Should I be using iodine instead?  What kinds of contams can live in 70% alcohol, do you have any links to references?

He was only speaking of tools you can flame sterilize. Alcohol on a clean paper towel is fine to wipe off an injection port

We wipe the SHIP with alcohol because we cannot flame sterilize it.

Alcohol does not kill mold spores on contact.....fire does.....

If we kill everything on the needle with fire and then wipe it with alcohol it then becomes unsterile.

Wiping the tools that cannot be flame sterilized with 70% alcohol is common practice, as it does sanitize.....but you could possible be wiping mold spores over everything.

Just expunge a few drops of spore solution to cool the needle after flaming, then the rest will flow contam free.

Good to know I've been doing it right not that I plan on using syringes much more. Agar boat here I am.
