HELP! What to do with my tubs? =/ Trich..

Hey guys I'm back with some updates with my grows and plans for the future for more =)
First off I have 2 buckets that have been colonizing for quite a while(1 qt spawn:4 qts coir/verm) and when I saw they looked pretty colonized I put both into fruiting conditions about 5 days ago so hopefully see some growth out of them in the near future.
I also spawned 2 more tubs with liners this time! At a 7:10 qt spawn/sub ratio on 12/21, which has colonized much more quickly than my first tub, already almost looks fully colonized but can't tell, I'll give it a couple more days before I mess with it though.

Anyways I'm going to have about a month of free time (no work) to begin some projects and would like to document and show you guys my progress along the way. I've bought nearly everything I can imagine for agar work now I just have to start something and get into habit of making agar and pouring it. I need to find another good source for large amounts of petris, message me if you know one maybe?

So my PDA mix says its approx. 20g dextrose 15g agar and 4g potato infusion. Do those measurements sound right because it sounds off to me :P Says the ph is 5.6 though and used for this stuff so it must work?!
My plan to make the agar was to mix 10g powder with 250ml of distilled water in the Erlenmeyer flask, microwave for a minute so it dissolves, then fill the top with polyfill, aluminum foil over that and pressure cook at 15 psi for 20 minutes.
After that how long should I let it cool off before pouring my plates in the SAB to avoid much condensation? I've also got parafilm to wrap the plates, I do that right after I finish pouring or after they completely cool?
I'd like to do my agar this Saturday because I have 1 qt jar of grains I'ma G2g to 7 and try to make one last B+ MS monotub before my agar work begins.

200 g Potato infusion [dilute to 1L total, ~4g solids] 20 g Dextrose 20 g Agar 1 L Distilled water thats what i've beeen using with great success or just 1/4 teaspoon of potato flakes
Pour your agar when it has cooled to the point where the container is still hot but can be handled and the agar is still liquefied.i also put my parafilm on after i inoculate looking good man keep it up agar is my favorite part! i have been using alot more pp5's for my work as of late to save my dishes for the later stages of isolating

I did some pour agar with tetra for some dirty ass prints and it is the fucking bomb. Then transfer to my pp5 for isolation Save the big guns for when there needed.

I see I see, thanks much I was confused seeing potato infusion in wet form rather than dry, but it should be fine (:
How long can I premake my dishes and still have good results using them? Like I just don't want them to dry out or something lol.
I found a great deal online for bulk coir bricks too, so I picked up 39 bricks for like 45 bux (no shipping), now I just need to go buy an asston of WBS and like 10 more tubs and I'll have something goin ;D
Now I just gotta figure out these large spawn bags and how to foldem up and put in my 23 qt presto. Hoping to fit 3 bags per cycle with 6 qts wbs in each bag..now that I think about it probably only 2 :P
Then I just have to start some masters off some agar wedges and let the experimenting begin

I dont understand what you guys mean? You have seperate kinds of petris and you switch them up for isolating? If that's the best way explain please and you'll be my best friend

Depending on how stiff you make your agar at least a month, probably more. But once you start agar, using in time won't be a problem, not having enough will more than likely be your issue (at least its mine).

well pasty does his bass ackwards from me but i use twist top glad containers and find it hard to see well enough when im going for iso's
 so i start my work in here and use real petris after a few transfers in pp5's

No pour agar is agar that is pre mixed and PC'd in the container. Something that can survive a PC cycle like a jar or pp5 plastic. The sleeves of plates are poured in a SAB from a flask after PCing. I use those for antibiotic agar simply because the antibiotic needs to be added when the agar is cooling, it will not survive a PC cycle.

Ya got me I'm just too cheap to do pour agar unless its for antibiotic purposes which is covered by health care

Yeah another reason I'm on the hunt for a cheap supply of bulk disposable petris (= I have like 30 now, but I'll fly through that I'm sure, just bought that to start, and EVERYTHING else for this hobby for like the next 2 years :PQuote:

And alright so because you start with so many genetics you rather use one of those containers to start with and make transfers from until its down to a few (since you can reuse those?) then switch over to petris in order to see and isolate further? =) Hopefully I'm understanding right.

you can use the pp5's through out the whole thing! thats whats great about them, another advantage is being able to put them on there side while you work giving you extra protection

Ah very nice, the side of the container will stop contams, just a bit harder to see :P I'll have to go make a purchase!
Must I use antibiotics in my agar for the petri dishes? What's the pros to using that, and where can I get some?

but yes the faster i can use them the faster i can re-use them and my iso's are kept alot longer so i use petri's for those

Not at all. I did it because I'm working with wild prints and they are dirty as fuck. Antibiotics will prevent bacteria from spreading and as molds can be easily isolated away from, it makes cleaning up dirty prints much easier. If your working with vendor syringes or prints its not necessary.
