Hello Folks, My first grow seems to be going well. The jars are very near colonized and hopefully early in the new year I can put them in a terrarium.
Fingers crossed the grow keeps going and I git me some fruit! I am planning to make spore prints when the time comes, but I read the LC cloning TEK:

Where you use a plug cut out of a mushroom stem as the genetics to inoculate a jar of nutrient. It's a fascinating idea, and might be the easiest way to step up production.
I had a few questions about the TEK. I was planning to recycle a leftover spore syringe to cut the plug out of the stem. Is that needle diameter big enough to work?
Also, is my understanding right on this--inoculating jars with mycellium results in very fast contamination because the genetics are already diploid, as opposed to using spores where they must first "find" each other, combine genetics, then grow?
It would be nice to inoculate jars with a healthy dose of mycellium LC, and watch them explode with growth! In my first grow, I was way too stingy with spore solution and they took forever to get moving. It worked out ok, but took at least an extra week.
Thanks for any LC thoughts--likes or dislikes on the technique.

yes, way faster with lc. I do lc,s some. but I use them right away. when I,m ready to start my spawn run to bulk sub, with fully colonized grain jars... I take a couple syringes of sterilized water. same tek as for making a syringe from a print. inject water, into my clean,fully colonized , grain jars. just like inoculation with flaming the needle ect. shake jars to break up some mycelium. tilt jars and draw out the lc. works good, and a pretty sterile tek....just my 2 cents on lc,s...have fun with it....

Thanks Blue--That's what I was thinking. The TEK said you can store them for around 6 months. But I was planning to inoculate a few jars (in case one or two of them get contaminated while I'm learning my technique). I was going to let them grow out for 4 or 5 days with magnetic stirring, and then knock up my grain jars with a heavy dose of the stuff.
That should let me grow out some shroom from the LC, and at the same time make spore prints from the nicest specimens.
How much solution do you use to inoculate a pint jar? I was thinking 1 or 2cc should really give them a heavy dose of genetics to get them to moving fast.

I still use this on occasion and yes that gauge needle works fine. If anything it helps fight contamination with quick colonization IME. 1cc is plenty you dont want to change the water content in the jar I do know some people have had trouble with this tek though. Good luck!!

Cloning to LC is very risky.

As long as your sample is contaminant free and your sterile procedures are up to par you will have little to no issues. But notahacker is right it is much easier to spot contamination on a 2d plane. also you can actually separate healthy mycelium from contamination with this method.

Thanks for the thoughts guys. I think I will try the technique just because--never can have too many tools in the box.
I do have a couple friends over in the biology department who could maybe give me a few grams of dextrose and malt extract. I see them bringing carts of this urine looking nutrient brother up and down the elevators all the time!
Anyway, I'll give it a try, assuming I can get these cakes to fruit. If it works out without contamination it would be a good way to get a quick second grow started. If it fails, I can dump it and go with spore prints (made into syringes).
