Ok so I still haven't isolated this specimen. Mainly because of how skeptical I am that it could be the AA+ spores growing. Let me run through the facts real quick.
- Out of 12 PDYA jar pertis & the 5 PBA petris only 1 has shown any type of positive growth, the one in question here in this thread.
- Inoculation occurred on 12/15 & the initial growth of said petri occurred on 12/21 (6 days after Inoc)
- Growth is white, but no rhizo growth yet after 3 days.
So those are the main concerns. Here are 3 new pictures of today's latest growth;

Should I still try to isolate this? Or wait longer? I mean seriously no other thing in that cabinet is growing anything that looks remotely close to that. Three of the PDYA jars look like it may be growing some bacteria.... that's about it.

Yeah... Just don't seem to be that safe. Or they are hella old or something. Hear all kinds of bad things about vendor syringes. Just gotta keep on keeping on till one day I get lucky. Hopefully one of these dishes grows myc, or one of my 4 cakes grows something I can work with.

Yeah looks bad, man. =frown
All mine so far have been from syringe, fwiw. So far 1 of 4 have contaminated with a bright green dot. The other 3 are all succeeding, touch wood. In all cases first growth was noticeable within three days, though really difficult to see as barely noticeable dots surrounded by completely clear rings of matte finish on the glossy agar surface only visible by viewing the glare from a lamp... if that makes sense. Two different syringes used for the sake of feeling like a bigshot. Keep at it, man.

So yeah, that thing is pretty much just looking more and more like cobweb, but in other news.
Check this shit out.
The other day when I used the rest of my syringe on some cakes I was getting ready to close up shop when I noticed a fucking ton of spores sitting in the cap of the thing like an hour after innocing.
So I thought to myself maybe I could do something with that. Not like I have much to lose. Looked at the petris on the shelf and thought y'know wtf ever, not like there's anything growing on these other things anyway and grabbed one of them(the cleanest of them all with no previous spores seen to the naked eye) stuck it in my SAB, opened the petri, opened cap to the syringe and banged the shit out of it to get the spores to come out.
It was not very safe, as a matter of fact I would say it was stupid as hell. banging out the spores with the lid of the petri so they would fall into the dish, but lo' and behold


Does that look like it should? Or was the cap just infested with more of whatever the hell was growing on my other dish. It's been three days since the spores hit that dish

Yeah that is too quick and don't look right to me, sorry. The growth is very random, even with a lot of spores, cube growth should be somewhat directional.

Don't pitch it just yet tho, there is a lot of spores on that plate, far more than I would ever use. Maybe its just cause there are so many that it looks so random. Give it another day and see how it looks then.

Word man thanks for the help. Just to be fair it was not sterile procedures at all other then a SAB.
I was very skeptical anything good would come from that one.
I followed my BRF cake PF TEK to a tee so all my hope is resting on one of my four cakes to colonize and pin so I can get something to work on agar with.
I'll have more posted tomorrow.

Agar is great for prints, but it sucks if you are using a liquid MSS. If I need to move from MSS to agar, I will make a plate or 2 of "brf agar". It takes the additional fluid much better than the traditional agar-agar.
Otherwise, odd looking growths at the edges of plates are generally not good.
