Questions about syringe viability and "sprouting" time of spores

Hello Folks,
I'm fairly new to this game, but have been reading like mad to catch up!

I had a few questions about spore syringes and also how substrate effects colonization time.

I ordered B  spores from a reputable supplier, and they threw in a Malabar syringe too.  So I PCed about 20 jars with the plan I would inoculate them half and half with the spores.

For the substrate, half the jars were mostly cooked brown rice, with about 1/4 Verm/rye flour.  To this I added some hot-steeped quinoa and whole flax seed.  These jars were airy from the coarse vermiculite and fluffed up rice.  Those inoculated with B  showed signs of life after just 5 days, and they are growing like mad.  The Malabars were a bust--none of the 5 showed anything after 2 weeks.

The other substrate was a standard 2:1:1  verm/water/rye flour.  All the B  showed signs of life after 6 days, but it's growing far slower than the wetter mix above.  Again, the Malabars were a bust---0 for 5.

I let them cool overnight, so I am certain I did not heat kill the Malabar jars.  Am I right to assume sometimes you get a dud?  I mean, it visibly had a good load of spores in it and I'm quite suprised they were 0 for 10. 

Also, what substrates tend to colonize the fastest?  It seems in my psuedo-scientific experiment that the wet, brown rice based substrate is doing far better than the drier Vermiculite based mix.

I don't really care about the speed of colonization, other than the fact that the sooner it gets colonized the sooner it can out perform contamination!

I used a 1/2" dry verm contamination barrier, and didn't suffer a single infected jar.  So for a first time attempt I think the technique and tools are sound.

Are their any rules of thumb about substrates?  It sounded to me like drier was better than having any standing water.  I guess I can see that--but my cooked brown rice jars are clearly wetter with a layer of condensation between the mycellium and the glass. 

Also, does it really matter what the grain mix is?  I've seen on here people using popcorn, birdseed, brown rice, whole rye....  Any reason you can't prepare those grains like normal (cooking or soaking or whatever), and mix them in any arbitrary amounts?

I inoculate organic rye berries because I can easily control initial moisture content.  I make my substrate using coco coir: 650g coco coir brick with two quarts vermiculite and 4 quarts water puts me at perfect field capacity. I then stuff the substrate into 7 quart size jars and properly pasturize that way  I will use a spawn ratio of 1:2 and fill my monotub at a depth of 4 inches. Works fine for me
