Questions about growth of my first grain jars with pics. *Update1-7-14 Continued grow log*

Hello, first time poster. Neewbie btw. I inoculated 5 grain jars last Saturday and still no signs(from what I can tell) of growth. I soaked WBS for 24 hours, rinsed before and after blah blah blah. Then simmered in big ass pot for 30 mins. Rinsed and let drain for another 30 mins. (Not sure if they drained well enough because strainer were small.) Into qt jars, polyfill used for filter. Then PCed for 80 mins. Let cool overnight. Then inoculated with MS syringe in a ghetto inoc chamber haha. They've been in my closet inside inoc chamber up until this point. Any thoughts would be appreciated. Thanks! Strain is Cubensis: KOH SS Samui

for future reference a ten minute simmer is all that is needed, if they were done right they should roll around like bb's in your jars and look dry. i've found the ksss(i know thats not wut you have) to benifit from a good consolidation and a casing layer, this reduces or even eliminates the short fat ugly fruits

I also have a PE-6 syringe and another B+ which I plan to innoc very soon. Used the KOH first for practice really. At this point the grains roll around like bbs which is good. Just wondering if I did everything correctly. Using room temp because I've heard good things. Pics are currently uploading btw.

Had trouble uploading pics. Gonna try again tomorrow at some point because I checked jars today. There was a few clump of grain conjoined by the mycelium. This was the case in most of my jars. So I'll just update it all at one time. Thanks again and Merry Christmas!

Ok pics thank goodness. Ok. Possible things I see wrong with the jars. 1.Possibly exploded too much of the grain. 2. Could have rinsed better because slight dirt residue on inside. 3. Not completely sure if water content is correct. No pooling of water at bottom and I know they're not dry so I'm confident in that area. 4. Too much grain in jars. Makes it harder to see growth with the excess grain. That's all I have speculated. Let me know you guys think. And as always thanks a lot!

It may be a little slower then normal due to the bursted kernals but next time..

One last tip I know its hard but I always heard don't handle the jars too much just leave them be for a week then check for "bad smells" then again every few days till shake time. Its more for the brf jars then wbs etc. but I just feel like its good practice in this hobby. Patience. GL

Christmas update: All jars have a few spots of growth! Unfortunately everything's closed today but first thing tomorrow I'm getting some rye and a few other things on my mycology shopping list. The box that contains the jars is in my closet. I've got a heater in there which keeps an automatic temp around 75. Hope all is well. Merry Christmas!

Ok so 3 out of 5 jars are showing decent growth. Very excited. Picked up 24 more qt jars yesterday with 2 big bags of wbs.(Couldn't find rye.) Along with a lot of other needed materials(strainer, mixing bowls, 2 60 qt tubs...etc.) Very throughly rinsed 10 qt jars worth of grain last night. Soaking in warm water. The plan is to simmer and jar those babies tonight when I get off work. Very excited!

                    NEW BATCH OF JARS 12/29/13
:mushroom2:Update: Dried simmered grain until toiler paper standards were good, lol. Fairly dry overall but still moist. Loaded up into 10 jars. Did a better job of not overfilling. :biggrin: Using pollyfill for filter/ge. Tinfoiled and then PCed 5 for 90 mins. Then later PCed the other 5. Both were successful. Grain has plenty room and rolls like bbs. :thumbup:
Plan to inoculate sometime tomorrow. I have 2 MS:syringe:. B+ and PE-6.

Question though. How much should I inject per jar. Both are 10cc. Basically can I spread a 10cc syringes over 10 jars. 1cc per jar ok?

Exactly the simmer is only to add water mainly, watch your grains...10 min tops...and rye is soooooo much easier...but good job and I hope it turns out...updates!!!

The only reason I have ever simmered grain was to get them hot enough to evaporate most of the external water.  About 6 months ago, I gave up the simmer.  I have had no issues at all with colonization of grain without a simmer.

I also found that with WBS i had a lot of grains exploding, once I switched to rye that became a thing of the past....

Eh. Not tryin to be mean or anything but still looks a bit wet to me. I could be wrong but I dont think u should usually get that many grains sticking to the glass with proper moisture content. They dont look overcooked to me though.

7 out of 10 lol

True true. They were just PCed 2 hours ago so they're still warm and it looks wetter than it will in a day or so I'm guessing. (I'm thinking the grains will re-absorb most of the extra moisture.) Thanks!

They also look a bit full to me. Just see how hard it is for you to break them up and evenly distribute the colonized grains again once you shake them. If its a bit of a pain than give yourself more room :smile:. When using ms inoculation the spore solution tends to collect at the bottom so that is where most of your mycellium is going to be for your first shake and getting colonized grains from the bottom of the jar to the top without room to shake can be a pisser.

I think most people fill master jars about 75%.

To answer your question on how much spore to use, most people told me betweent 1 and 2 cc per jar is good. I use two but really you could do it with any amount theoretically. Just take a bit longer since you have a smaller amount of inoculate.

Ahhh ok yeah. My grains always look wetter right after pc aswell. Your probably spot on then.

You should only shake once at 20%-30% colonization. No matter inoculation method or species.

With spore syringes more is not better. The more solution the more contams get introduced.
That's why isolation on agar is beneficial.

Didnt say better, said faster/slower :smile:. Hopefully he made/baught syringes that were properly produced in a sterile environment with distilled water so there should be no contams but ya never know when you dont do it yourself.

Also dont forget about the performance benefits wich is the true reason behind doing agar. You can isolate a fast colonizing/large fruiting section. With ms you never know what your going to get for genetics. I just went through a six pack of syringes from a sponsor shop and I swear five of them were friggin junk. I mean they all worked just crap genetics, taking two friggin months to colonize a jar, wich isnt the shops fault. Just bad luck. Thats why agar is my next step :smile:

also like to point out that you should only HAVE TO shake a jar once at 20-30% colonization. Not that you shouldnt do it again if it becomes necessary. Like if it stalls out or something. Just a clarification so no one gets confused :smile:

I give my grain a day or 2 to allow for the moisture to stabilize, but I think your grain jars are too full.  It can make shaking hard when they are that filled.

From the pictures it definitely looks as if they're too full but I'm pretty sure they'll be ok. I'm going to give them a day or two. Then inoculate. I'll update with new pics soon to see if the moisture content has stabilized. Thanks again!

Ok. So I gave the new PCed jars some time to cool down and get they're shit straight. I gave them a firm talkin to and an intense shakin.:warcry: They seemed to start acting right.

But in all seriousness.I'd have to say the jars look exceptional! As always all opinions are appreciated. :rockon:

And for all of that "too full" talk. Pfft. That's a thing of the past! Lol :suckit::harhar:
I'm planning on impregnating these 10 jars with one syringe of PE-6!

that little space heater mite dry out your grains. jars colonize quit well just sitting on a shelf at room temp. remember its abuot 5 degrese hotter in the jars than outside. so don't over heat them.

Ok, interesting point. But the heater has kept it consistently at 75F for an entire week now. Is an ambient temp of 75 too warm?
As for drying out the grain jars, the humidity is relatively high inside the "incubator" box.

So I'd have to disagree :wink:  but I really appreciate the feedback! Any and all critiques are encouraged! :rockon:

75 is a bit high i think. Giving that temps inside the jar are higher and mycellial growth rate slows at 80f. I keep my room at 70 and seem to get the best growth at it. For cubes anyways.

Also if you can atleast turn that heater so that it doesnt blow directly at any tubs or anything it might be good. Once you got monos in there a heater blowing at them is going to fuck your fae and humidity. Thats why tc's say to use radiant heaters but I use a ceramic myself aswell. Just try not to have the air current from it blowing too strong on anything.

There that one counts cronicr :smile:. You can go back to answering everythign again lol.

I beleive optimal temp for fruiting mushrooms is between 74-78f (according to paul stamets mushroom cultivator). Im not sure about other stages. I just go with 70f for colonization of jars cause over the course of six grows ive tried different temps and that seems to be the sweet spot for me where I get the fastest growth.

Also keep in mind ofcourse, as previously stated, that temps inside your jars AND tubs will be higher than the ambient room temp. So if you want 75 in your jar, you want ambient 70f

This information is also for cubes. Other varieties may differ.

Higher colonizing temps just make it easier for contams to form and costs more to heat. I have an oil radiator heater (better for mycology as it doesnt dry the air out as much as what you have) and I keep it in my room with my closet door open, temp at 75 (goes from 72-79 depending if on or off, automatically) and everything I have has been great!

75 is fine imo

I am not quoting you as a specific individual, but for the information you and others provided.  This is not the first time this rumor popped up.

Higher temps do accelerate the growth of contamination. Higher temps however not create nor form contamination.  They have to be introduced.  If you make a cake or grain totally sterile, there is no contamination.  IF there is a contamination, procedures were lax.  Contaminates do not just show up with it gets warm. 

Its a matter of perspective.  People set their jars at higher temps, and noticed that contaminants growths were accelerated. I would wager that most MSS syringes are simply not clean.

I can almost guarantee that If I take one of my jars that was inoculated with agar, and increase external temps to 85-90, I doubt I would get more than a stress reaction from the mycelium.  I have no way to test it, nor would I. 

We are working with totally sterile media at times.  By the flip side, it is VERY easy to introduce a contamination, and most would love to be in a place with lots of food, and no competition.

This is just my impression of the contams/heat relationship.  I may be totally off also.

Everyone says to shake grain jars at 30% blah blah blah. I understand this. Should I use some surface area calculations I learned from middle school?  :justdontknow:
How do I know when jars have reached this point. Again you'll probably be able to tell from pics but just for future reference.

Ok so 4 days later on Tuesday 12/31/12 (New Years Eve mycelium babaayy!) :whoo:



As you can see from the first pic I provided the humidity inside the tub is relatively high!
Also I dropped the temp a bit just for experimental purposes/whythehellnot.
The last pic is of one of the jars. A big clump of wbs has formed at the top. It had sweat inside of jars so I'm thinking that it's created a myc ball? Lol anyways Happy New Years to everyone and I hope we all have a great year of mycology! Cheers!  :bigblunt:  :rockon:  :toast:

So far it looks great.

Last month, I started testing every other agar xfer....which Has pretty much fucked me for time (I will elate)

Half of my jars I have been trying to get the wedge of agar down one side.  The other half, I am doing the same...followed by violent shakes.  My goal was to obliterate the agar wedge.  Most jars are 100% after 10-15 days without another shake.  Meanwhile the jars that were not agitated hit 20%.

looking good so far. your new lower temp should keep the internal temp in your jars right in the middle of the optimum range.

Like letting the substrate fully consolidate which gives big pinsets and such ei. a lot of primordia and knots which equals a lot of pins?. 6500K lights? best way to raise humidity?  I'm sorta trying to make a list lol.

bulk subs don't need tobe consolidated you can fruit right at 100 percent but if you do 3 days is enough,all ste ups from sg's to gh's to mono's are capabble of the wall to wall the right genetics key, just follow your pinning triggers and you will be fine either way you choose

Just to make sure were on the same page here, there is a difference between colonization and consolidation. When your sub is still brown and being taken over my mycelium that is colonization. When its 100% white and you still let it sit for a while that's called consolidation. Just wanted to make sure. Plainest way that I can understand it is that the mycellium sorta recovers its str and does some body building while it consolidates before reapplying itself to fruiting. This generally isn't needed with cubes cause there such a robust species already. Same reason that casing layers aren't needed.

Other than that, yeah all forms of fruiting chambers are great and can, as cronicr said, give you the pretty wall to walls :smile:. That being said most major cultivators use mono's for the lion share of there work. There the easiest for real bulk.

Controlling humidity: Three big things on that are proper feild capacity of substrate, re hydration of grains and oil filled radiant heat if any additional heat source is needed (nothing with a fan creating air current). If that's all good than a lot of the time your tubs will care for them selves really. If anything more is needed just mist the sides of the walls and Ive heard a lot of people (im trying it myself now) use a thin layer of vermiculite on the top as a casing layer and mist that as-well. Reason being that it changes color when it dries so its a easy visual cue. With sgfc's you can also redunk the perlite between flushes if needed :smile:.

Awesome response bro. Yea my basic understanding is the longer you wait before fruiting the better the pinset will be because it had more time to consolidate. More knots and primordia and then trigger pinning = wall to wall hehe. Also what would you guys say your average or your typical dry weight of flushes from a monotub. I know there are a ton of deciding factors but I just want to hear some numbers from personal experience. :takingnotes:

The major pinning triggers are in order of importance, full colonization, a decrease in CO2 levels due to increased air exchange(not gas exchange which is minimal), a steady rate of evaporation from the substrate or casing layer, and lastly, light.

Hyphal knots form best in 100% humidity, but I didn't list that because it's not a pinning trigger, but rather an environmental condition that is necessary. That's why we use casing layers. The casing helps to provide the 100% humidity right at the surface of the substrate where the hyphal knots form.

I have seen no correlation with temperature drop whatsoever. In the summer, my growing chambers are 10 or more degrees warmer than the open shelves I incubate on due to the heating effects of the lights. Even with a temperature increase, I still get wall to wall pinsets, so I don't consider temp drop relevant at all to tropical species. Other growers disagree of course, but that's just my observation after many years.

Full colonization of the substrate is the number 1 pinning trigger. Full colonization can be when the mycelium reaches the physical border of the container they are in, or when they run up against a biological border, such as a contaminant species. Either way, they see they have colonized all of what is available to them, so they then enter the next phase, which is reproduction.

There must be evaporation of moisture from the substrate for pins to form. A waterlogged substrate will just sit there forever without pinning. Even in 99% humidity, as long as you provide fresh air, moisture will be evaporating away from the substrate, and this is necessary for pinning. We mist to replenish the lost moisture, then allow it to dry slightly before misting again. This keeps the moisture content high, and keeps the humidity at the casing surface near 100%, but at the same time provides the evaporation of moisture that is a very important pinning trigger.

During colonization, we provide very small holes in the jars or tubs for gas exchange. We want a high CO2 environment during colonization, because this prevents the mycelium from consuming all of the substrate. The mycelium colonizes the substrate, but doesn't 'eat it all up' due to the high CO2 levels. During fruiting, we remove the covers to provide air exchange, which is at a much higher level then the minimal gas exchange provided during colonization. This increase in air exchange lowers the CO2 levels, and is a major pinning trigger. At this time, the mycelium begins to consume the substrate it has previously colonized, and we notice during fruiting that our substrates pull away from the sides of the container. This is not due to moisture loss, but rather due to the mycelium 'eating' the substrate and turning it into CO2, a waste product. It is easily proved that this shrinking isn't related to moisture loss, because even when we dunk a bulk substrate, it doesn't return to its pre-flush size.

Last, but not by any means least is exposure to light. Light does much more than just tell the mushrooms which way to grow. There are mechanisms in the light that stimulate the formation of hyphal knots as well, and light at the higher end of the spectrum(blue) definitely, absolutely stimulate more hyphal knots(which grow into primordia, which then morph into pins) than light at the lower end of the spectrum(red) This does not mean to get a 'mood light' with a blue lens, but rather to select lights such as metal halide, or much more economical is 'natural daylight' fluorescent that emit light at around 6,000 kelvin to 7,500 kelvin depending on the brand. Cool white fluorescent emit light at around 5,000 kelvin and the 'red' incandescents emit light at around 3,000 kelvin. The higher the light temperature in kelvin, the more stimulatory it is to hyphal knot formation. I hope this helps.

RR is the fucking man lol. He just has the way of speaking that breaks everything right down to its fundamentals and explaining so a child could get it.

There trully are just so many variances in play that to try and count on any particular weight of return on flushes would be an excersize in self defeating futility. That being said...

Starting from a mss with atleast average genetics and giving that all your grain prep and pasteurization etc was done correctly, ive experienced an average return of 4-5oz dry weight from first flush on 60 quart tubs receiving 6qts spawn and 10qts sub of a basic nature (coir verm thats it). Just seems to be whats average for me.

However, just to give you a look at the other side of the fence... FH has stated that with his recipes for grain and sub prep (including things like coffee and gypsum) and using isolates of his own he can average about 8-9oz dry weight from the same size tub and spawn/sub ratio.

And PLEASE do keep in mind to not count on or get your hopes overly up for anykind of return as this hobby, especially when working with ms, is just unpredictable and therefor uncontrollable. Any number of things from bad genetics to contamination to human error to mother fucking nature can decide to bitch slap you at anytime lol.

As promised I made my sab with a coffee can:lol after I got off work tonight.  I inoculated the 10 jars with 10cc of PE6. Pics will tell the rest. :whoo:


First time flame sterilizing question. I got it red hot and then waited 5-10. Squirted a little out and then inoculated. It just kind of scared me because the end was hissing because of the fluid at the tip of needle.(during the flame part :lol:)
At one point I somewhat quickly inoculated a few jars but on RR's videos he says it doesn't even matter. ("the fluids just gonna cool it anyways") How long should I wait? I'm just making sure and just wanna hear some opinions as usual.:takingnotes: Just wondering. Thanks as always! :rockon:

3 out of 5 are showing great growth. Do you think the first jar(pic 1&2)is ready to be shaken? And as always I have a "special" jar.
This is when the mycelium in a jar decides to break away from the crowd and grow in a different fashion. :lol:

MY OH MY OH MY. I picked up jar #1 and began to shake it like a polaroid pictchaaaa and instantly my nose was filled with this gorgeous, pungent, odor of mushroom.
I've never experience that before and it was absolutely breath taking.
Lol. All jokes aside I shook the fuck out of it and beat it on my shoe until the big clumps were broken up & spread evenly.
The odor was just so strong and it smelled like straight mushroom smell. :mushroom2:

2 DAYS AFTER SHAKING
Blown away with the huge amount of growth. Couldn't look any better in my opinion. Hehe.:whoo: I guess after I gave em that hard shaking they just woke up and was like "we got some growin to do!" lol.

It has been extremely cold in my area for the past week. Like 0F and below. I have no central heating in my current household and therefore rely on space heaters to keep my room warm. They do a fantastic job and my jars would have to agree!

break it up. it should have been wayyy more profuse than that by now. Shake it and see if it recovers, but just a heads up there may be soemthing wrong.

Yea I know the problem. Jars don't have proper moisture content and grain is too exploded. Newest PCed jars are perfect tho hehe.

i'll never argue with a g2g:shrug: spawn one to 7 or however many your pc holds and once your sure there fine spawn the other 2 to trays(or more g2g!)

Or you could spawn all three to like 12 more, doing about a quarter of a jar to each new jar. More spawn to each jar means more initial points of colonization, leading to faster finish times. Plus 6qts of spawn to 12qts of substrate gives me right about 4" substrate depth on my 60qt tubs. This is assuming you do the standard 1:2 spawn/sub ratio on your tubs.

Im actually thinking of doing this with a portion of my current grow. Started from mss's and got 11 good colonizers and 5 not so good. Spawned 8 of the good ones and thinkin I might fridge the remaining three till I can pick up more seed and try my first 10 jar g2g instead of spawning em with the crap five. Be very interesting to see 30 jars of 100% lol.

Tomorrow will make a week since I inoculated my prized grain jars. I have seen no signs of growth as of yet. I had the syringe since late October'13. But kept it in a cool place so I doubt it was a bad syringe. Well trusted vendor.

You can get away with it but most people would reccommend atleast a 1:2 ratio and a 3-4 inch substrate depth. So depending on your size tub....

Will do. Got the 10 PE6 on the way.( I hope) Gonna get some better bird seed. More millet, less corn. Innoc with B+.

Just to clarify cause I'm a noob. I can spawn 4 qts of grain to 12 "parts" of substrate. In 60qt tub? Thanks alot. I searched but couldn't find easy noob info. :toast:

I would maybe try a smaller tub or using something to divide that one. A 60qt tub (wich is what I use) takes about 21qts to create a 4" substrate depth. Thats the recommended depth for the best returns and to keep substrate drying rate controlled.

Also a 1:2 ratio wich is recommended would be 4 spawn jars to 8 sub jars.

Again you can go macgyver it anywich way you choose but these are the suggested and I would recomemnd trying to stick to them atleast to start out. They are the norm for a reason :smile:

EDIT: Im not trying to nay say or anything like that sir. I am just trying to share from experience. A lower spawn ratio is going to cause you significantly more colonization time. Now in jars it takes what it takes whatever for the most part cause those are sterile so who cares. But with sub it is just pasteurized and its just a matter of TIME before a contamination sets in. I have done exactly what you are asking before. 3 times in fact and I got 1 flush from one attempt. Everything else was lost to contamination. You want to storm your beeches with asmany troops as you possibly can to assure victory.

beaches being substrate and troops being spawn obviously.

Please learn from me so my mistakes were atleast not in vain lol.

EDIT2: Even if you just like tape some parafilm up to divide your tub in half. That would be just about perfect substrate depth with 4 jars spawn and 8 jars sub.

yeah 1:2 is normal up to 1:4 is acceptable, if your gonna go with the four make sure you dunk your grains before you spawn, that will help your colonizing speed, for future refrence 1:2 may get you more flushes

The moisture is off your grains, nothing to do with any kind of growth really. Looks like you have a TON of cracked corn in there though, I'd look for a better brand of wbs next time, and the moisture really shouldnt hurt you, but you can always blowdry your grains before you load them into jars to prevent that.

I add corn to my WBS. I find it makes things easier to break up later on for spawning to bulk or G2G. I also blow dry my grains on occasion.

Idk bro. Grains had pretty good moisture content even after PC. Let them sit for a week and then inoculated. I doubt a random bit of moisture appeared at the top. All I'm saying is when I see mycelium growth starting there is always patch of sweat on the glass near growth. Idk. Maybe you're right but idk. Ps I did blow dry them lol

Actually it would make sense. As the mycellium colonizes it creates heat making the insides of your jars a few degrees higher than your ambient temp. Thusly the sweat.

i would say just the moisture in your grain evaporating, creating this moisture. you should see small spots of myc against the jar if its growing. in my experience...gl

Depends on how the jars were done. If the inoculation media was placed in the center you will have growth before you see it. Thats why usually teks will tell you to spray your syringes on the sides next to glass. Also if the jars sat for a week before inoculation without showing signs of sweating on the jars than something had to change to cause it.

about a 30 qt tub would be perfect, you certainly can case but it's not needed, helps if your ambient rh is low though

Here my tub. Dunked the 4 qts of grain before spawning. 12 qts of sub. 1:3 ratio is ok. 3+ inches of sub total :headbanger:

No holes as of yet but that's ok. First mono so I'm just gonna drill 'em when it's time to fruit! Critiques and criticism is encouraged! Thank you all for your help up to this point! :thanx:

Probably gonna make a new thread for the tub but the 4th jars was thrown in with the other 3 for a better spawn ratio.
I've got 10 jars of PE6 coming along nicely with some good sized growth atm.

PE6 texan. From what I understand is a cross done by RR of PE and a texan strain. Something like than anyway. T.H.E. is the place to go
