MYEA plate prep question
    #19294210 - 12/18/13 05:02 PM (10 years, 1 month ago)

My bac-t background only ever had me preparing TSA, MP2, TNT, and MacConkey plates, and I've never worked with MYEA or PDA before.

I ordered (from a Shromery preferred vendor) some pre-poured of each to start with, but I also wanted to prep and pour my own so I bought a premeasured pouch of MYEA mix.

Now, in my knowledge of plate preparation, it was always

1) Measure your di-water (in this case, 1L)
2) Decant to appropriate sized Erl flask on a heated stir plate
3) Bring water to stirring low boil and add powdered ingredients
4) Continue heated stir until all ingredients are dissolved.
5) Sterilize
6) Allow to cool, pour plates and allow to set

Now, it's step 4 I'm concerned about.  I brought the water to a low boil and stirring for 10 minutes still resulted in some light sediment in the flask.  Another 10 minutes with additional heat, and the same result.  So it seems that there was just something in this premeasured mix which wasn't going into solution.
So, I distributed the mixture to smaller containers which fit in my PC, and sterilized appropriately.  Afterwards, there was still that light bit of sedimentation.

I let things cool, prepped my materials and SAB, and poured my plates, making sure not to decant the sedimented material.  The plates are currently cooling and I'm hoping they'll A) set properly, and B) grow things well.

Is this common with MYEA, to have light sedimentation remaining?  I'll keep things updated with plate progress in this thread, but right now I have my doubts regarding this batch.

Man, that sounds like a fun, and good paying job. As for the MYEA it is probably the yeast, I have the same problem, but, and it looks as if we both signed up at around the same time, so I am fairly a noob myself, except it sounds as if you have worked in this field(I would love to do something similar) but I am a construction worker, anyway from what I have read its the type of yeast that is used, other than not being able to see what you got going on underneath( as in holding up to light and trying to see the mycellium on the plates top) I have not encountered (as far as I am aware of) any negative drawbacks to this here is a pic of places I get stuck working in, and I am deathly afraid of heights, but I have a family to take care of and it does a good job of paying the bills(except the last 2years have been badd)i have used everything for replacement Petris from condiment cups to dq sundae containers, I love making agar! The funny thing is almost always the Petris will cont. when they hit the edge, the others worked fine, just to bulky! As you can see the agar in the dq containers are all diff. Colors I guess this was from not stirring as I poured having an unequal amounts of Nutes , but in the end everything was ok.

I usually get some light sedimentation when pouring my LME plates.  (I'm pretty new to agar work though). Heating and stirring (about 10min) before PCing definitely helps, and I try to mix the agar up in the bottle after every couple of pours or so in the SAB.

If I let it settle and gel in the agar bottle without swishing, I can definitely see a color gradient in the cooled agar.
