2nd xfer from pe mother (no-pour) dish. I would like one more clean transfer and a reduction in genetics. This jar used a very nutritious agar, and after it enveloped the surface, began to make pins. None got bigger than that, but 8 were xfered this evening. Using agave nectar and karo agar recipes in hopes getting this process moving.

after not being home for 5 days i came home to all my dishes fully contaminated so i'm gonna noc some more up today and hopefully be able to be more diligent about my sterile technique and watching for the nasties...

No-pour? I do no-pour and found that 20 minutes in the cooker is simply not enough. 45 minutes works 100%. I am not certain your method however.

No i do a pour in a SAB i was thinking about switching to no-pour but i have a bunch of dishes already poured so i just used those. i do my agar in the pc for at least 40 min. My failure with my first inoculation was my sterile technique coupled with wild spores and not being home for almost a week to catch the bacteria that had moved in. i have had no problems with clones, grains or transfers on agar before i just was a little distracted and not home, but as soon as i run out of dishes i have the pp5 ziploc half pint jars already so i'm definitely gonna try it at some point. i'm glad you're here whippy you seem to be very knowledgeable at this hobby called mycology!

 this is my 2nd PE transfer onto PDYA from a leftover grain dropped doing g2g in my SAB that grain master had a full canopy and beautiful fruits!

I had initially cultured PE from a swab to a pair of nute heavy dishes. The slow growth from the nutes and the apparent overage of spores created something that I simply did not believe was even a mushroom. The swab was tossed out right after, and one of the jars was retired for being too old.
I moved a number of what looked like pins forming to a few faster growing media. I hope to get some good rizo growth, get a small spattering of genes and send it off to the grain factory.

That bugger has growth all over. Its bonkers how one side is so hungry, and the other is simply content to sit and digest. I grew out a grain one time. It was a great time, and I figured that a millet seed being so small shouldn't have too many genetics.....woah, I think it had all of them. I think I ended up keeping track of about 30 jars, differing poor-man-agar recipes, and never fruited a single one. I stumbled upon a very large fruit that needed cloned, and my studies took a different direction. I learned quite a bit about nutrients and benefits on agar because I refuse to order supplies. I have used quite a bit of at home stuff that has and will work well for different purposes.

i meant to get a clone off the tub i had done of PE, but i was harvesting right before and after work that weekend and never had a chance to keep up those 2 tubs came in at over 3/4 lbs. from a single grain transfer from an MS, first flush. this time i only grew out one tub so hopefully i'll get a nice clone! i don't order any of my agar online either i like to have complete control of what goes in everything i do is local and made in my home

I re inculcated my petri dishes yesterday. Hopefully I'll pay closer attention this time... I also spawned some grain jars to straw hpoo coffee buffalo poo ewc and gypsum.

I set up a race for 8 transferred cultures the night before last.
All PE transfers are showing evidence of recovery. I am very gentle with moving cultures around. I am hoping they decide to go fast to the edges.
I had also made a few xfers from a more recent MS Eq plate. This was an indoor print I collected.
I cut back nutes in the agar on a few to really get them jumping.

Just got back in town from a Xmas party and got germination on my wild Aussies, no visible contams Looks like my plate with the hericium coralloides is doing alright with no visible contams and that print was guaranteed filthy hopefully I see germination soon. So far the tetra agar is doing its job.

100% of Pe xfers failed. Each picked up an unidentified mold. I had to chuckle. In 3 months of using the SAB, I have yet to pollute anything.
2 Survivors (not PE) are alive and well. Leads me to think the pe was a fail, and it is being relocated to the dead wood pile.

Aussie were incredibly tomentose from germination. They are now on there 3rd transfer and starting to look a little better. I am thinking about isolating 6 or so and running a bunch of little tubs side by side.
No growth yet from my purple mystic, hopefully I see germination soon. Made a few transfers from my PE and am hoping to see sectors soon there. Just put some pan cyan Jamaican on agar yesterday to so we shall see how they go.
On a non active note I also got some hericium coralloides and some shaggy mane wild spores on the tetra PDA as well, hope to see something from them soon too
