Will they breath and thrive (10) inside a closed cardboard box?

they need Gas exchange. so the small holes at the top of the jars would be enough for colonization. mind you it has been proven that mushrooms benefit from light exposure in all stages of growth, so you may have better results if you expose them.

I put a book on mine and let it breath through the cracks not covered.

Could light exposure during colonization affect potency?

My jars colonized in 20-23 days, ( some of the noc up points didn't get spores so a few took their time to get around) and were exposed to very little light. Could be the variety I got, or the fact the vendor sent a syringe with a massive glob of spores in it!

potency is a genetic trait, so i don't think exposure to light will make them more potent. but it may increase your total yield or speed of colonization.

I've read that longer consolidation times can increase potency dramatically.

The only difference I've noticed is those colonized in light seem to start pinning more readily.

i've seen no study to support this. most potency discussions are based on hearsay and speculation.
i would say that genetics of the specific strain of mushroom you are growing is what determines potency ( by strain i mean the genetics created when two spores combine and create clamp connection.)
