Heavy bacterial contamination

What's up with your procedures?

My first question would be how you prepared the grain.

Second would concern your inoculation method.

:whathesaid:

They look like they might've been too wet after the prep. Any pics of the prep itself?

Bacteria for sure.

It sucks, but shit happens. There's no saving those. toss them out and try again.

Followed this tek to the letter. Had success before but now the liquid. Literally followed Magesh's tek to the letter with the ezception of using a syringe bought from thehawkseye which I have used many times before and have known others to use. Knocked them up thru a tyvek filter put the foil over the tyvek and put them in the incub

The bacteria isn't a matter of grain preparation.

Somewhere along the line, the contamination was introduced.

Edit: Though if your grain is too wet, it will contribute to any introduced or already present bacteria.

I inject thru a hole in the lid covored in tyvek then out foil over the top of the jar and incubate. All surfaces and the air are sterilized well

So, you have a hole in the tyvek when you're done inoculating? If so, that's likely the problem.

The surface is not sterile once it is in open air. If you didn't flame sterilize your needle, then you introduced contamination that way. If there is a hole in your tyvek, the contamiation could have gotten in at any point in time.

when i used syringes i used polyfil, inject right threw it, no need for foil on your jars they need to breath

:whathesaid: ... use polyfill or SFD's for your lids. If you gotta use tyvek, atleast cover the hole with a breathable tape

@Wildernessjunkie

If you don't think you can get bacteria from how you prep your grain...
try PCing straight out of the soak water.

Oh wait you made an edit

These grain lids work like a charm. I personally use the higher end self healing injection ports for inoculating grain with MS instead of the blob of silicone but both work great. Follow that lid tek and get ready for success.

I flame the needle every single injection. I used to put bandaids over the injection holes but got told itwas pointless. I know nothing of polyfill

Yes. I edited. But Ill still disagree on this point.

When you PC at 15 psi for 90-120 minutes, it should kill everything in there. While there are a few things that can survive such temps, they are few and far between.

Assuming proper sterilization procedures, the primary cause of contamination is an introduction by the person inoculating the grain/substrate/agar/whatever else.

This is especially true if working with spores. Spores are not sterile. Aseptic, yes. Clean, maybe. But one thing is for sure, they are definitely a contamination vector.

Glad I could give ya a laugh friend, when i read polyfill i thought "maybe it is something like FA or RH another term for something" i worked in a pillow factory for a few years as a teenager, I know that kind of polyfill, still glad ya got a laugh from it

I was putting foil on because I have always been under the impression it was a standard thing no matter what, plus I have tons of the stuff everywhere, i always PC it on the jars. Also I was always taught to just cut a tyvek envelope from the USPS to fit the jar lid and and inoculate thru it and put a bandaid over the knock up hole, like i said earlier i got told i was pissing away money and time with the bandaid.

Heard.

It's always a good idea to put foil over your jars when you PC.
That way your filter won't get wet & wick contams in your jar.

I've never used tyvek as a jar filter personally. If I was in your shoes,
I'd go buy a cheap pillow and use the shit out of there as your GE filter.

I like the SFD's cause I don't have to change them every time I do a new spawn run,
but they are a little pricey. Def worth it IMO
