Shiitake spores relevant?
    #19225021 - 12/03/13 11:50 PM (10 years, 1 month ago)

I just noticed some shiitake spores I transferred finally germinated.
Is it worth trying to fruit this myc?
Should I work on isolation?
It took several weeks to grow and I'm not sure if there are even two spores to create fruiting myc. But it does appear clean

You will probably want to isolate a section of it and look for clamp connection first to verify that it is dikaryotic.

Kinda hard to see in this pic, but they should be at the septum.

No scope so I might just do a trial run in a small bag. But are the chances good that I have a decent strain here?

Well, you can kind of  tell the difference between them from macroscopic observations, but it isn't nearly as reliable. These are shiitake's as well, the faster growing thinner myc is monokaryotic and the denser, thicker slower growing is dikaryotic.

I've often found it really hard to see clamp connections, even at 1000 power oil immersion. The photo above is from an electron microscope!

Lol I'll pull out my back room electron microscope and check... Oh hang on...

id isolate all the germination points and give them a run if you have the time.

It seems I have a healthy dikaryotic mycelium. My dish looks like the second dish bottom right

This one fruited straight off the agar, but it was shiitake 75 and just in a mason jar:

If all else fails you can probably just clone some from the store.

I do have a knight velvet slant but these spores seem much healthier once transferred to agar
