Pink Contam Even After Pressure Cooking? [BRF]

Hi everyone! I am following PF Tek for P.Cubensis "B+ strain". I started a little over a month ago, prepared 4 1/2 pint BRF jars (1:1:2 ratio, although i added too much water this batch), used dry verm layer of course, covered with foil. I attempted a half-ass fractional sterilization (boiled jars 90mins for 2 consecutive days instead of 4). Long story short, all jars were hit with pink contamination.

I buried them outside for the hell of it.

Round 2:
Sterilization was key. I went out, purchased a Presto Pressure Cooker (rocker top w/ 15 psi), a can of Lysol, %70 Rubbing Alcohol, Latex gloves and disposable face masks. I prepared 4 BRF 1/2 pint jars (again 1:1:2 ratio, this time with the correct amount of water) & used dry verm layer, covered with foil, no tape. I PC'd the jars about 60 minutes (started timing when rocker began to jiggle.) I let them cool for several hours, then prepared room for jar inoculation. I sprayed plenty of lysol, put gloves and a mask on, dumped rubbing alcohol on gloves and rubbed together. I then proceeded to inoculate the jars, flaming the tip in between jars. (I do not feel comfortable with this however, and will be using rubbing alcohol and cotton to swipe needle in between jars instead.) After inoculation, i placed in a specially designated cabinet that has a large tupperware container in it. The cabinet was sprayed with lysol (before and after) and even took the liberty of wiping the tupperware with rubbing alcohol beforehand. The tupperware was never sealed, but the cabinet was (although not completely, as I have a heat pad resting under the tupperware that the jars sit in and the cord prevents me from closing it all the way.) Every time the cabinet is opened for inspection, lysol is sprayed inside. The heat pad pretty much sucks which is why I decided to use it in the first place, it never gets too hot. I have a digital thermometer placed in the cabinet, and reads anywhere from 75-85°F (Had it reach close to 90°F once, quickly addressed situation) with cabinet humidity being around 30-45%. Mycelium growth was more rigorous than the first round, due to less saturation and the addition of the heat pad. I decided to take 2 of the 4 jars with me and leave 2 jars at my friends (which is where I am primarily growing them). I only did this so I could monitor the jars myself and get an idea how the others might be coming along. I was skeptical about this as my house is a good 45 minutes away, I felt I was risking contamination during transportation especially at its current stage being uncolonized completely. A few days ago, I spotted that exact same pink contam in the jars I brought to my house.


I quickly called my friend and asked about them, and my friend reported that 1 of the jars did indeed have a pink spot but the other supposedly has absolutely no sign of contamination. I think it's just a matter of time IMHO...

I need someone to help shed some light on what exactly I can do to improve my sterilization techniques (preferably without building an inoculation chamber but if I HAVE to, I will). Is it possible that my spore syringe is contaminated? I did order 2 syringes, and have only used 1 so far. I also am unsure as to whether I flamed/wiped the needle after the LAST jar inoculation the first failed round. Perhaps contamination made its way up my syringe? Looking for 2 cents! :smile:

- Make a controlled jar (not inoculating 1 jar after PC)

short and unhelpful answer is it could be caused by any of the factors you sited.

There's a ton of sterile technique info here start reading and cross checking with your own procedure.

my 2 cents: make a SAB, soak a dishcloth and lay it at the bottom, put everything in you intend to use, close it up, wait for air particles to settle on to the wet dishcloth (anywhere up to 40mins). Spray gloves, insert into the SAB slowly, do work, finish up have a cider. Good luck.

Are you using the same contaminated spores you inoculated the first batch with?

I know you what you are telling me to do, and I've seen a topic about it, but what does SAB stand for? I've only heard it called inoculation chamber or a glove box. Thank you!

Let's just assume I did not sterilize, although i did not draw back on the syringe, could bacteria contam the entire syringe?
I did however draw back the syringe before I started to create air room to shake up my clumped spores. Should I have done this under a flame?

That's a possibility, you don't need air in the syringe to shake it. I don't know if it's necessary to flame the needle just before you put it away but I always have. It vaporizes the remaining water in the needle part.
