Golden Teacher Mono Tub * 1
    #19210787 - 12/01/13 12:20 AM (10 years, 1 month ago)

This is my first time working with Golden Teacher.  It's a cubensis variety that I have always wanted to research.  I have been holding on to two GT prints for a little while but I never started a culture because I only had two prints and I didn't want to make a syringe to find out if the print was or wasn't clean at the expense of my print being a dirty print that I lost due to contamination.  So I wanted until I was comfortable with agar work.

Once I had some experience with and was confident that I could successfully work with agar I grabbed one of my two Golden Teacher prints and inoculated an agar dish in my glove box.  I transferred the original culture a total of three times.  My final culture was not a mono culture, but once I was sure that I was working with a clean culture I inoculated a light malt extract liquid culture with a small wedge of agar in my glove box.  I also transferred a small piece of agar to a new agar dish so that I would have something to fall back on in case this project goes bad.

Since making the LC and making the fourth culture transfer I have inoculated a grain jar with a wedge from the dish which I will be using as a master grain jar to be used in my first ever grain to grain work.  But that's enough about my master grain jar because that will be a new post altogether once that jar has reached 100% colonization.  Now back to the LC that I made with a wedge from the third agar dish.

I think Paul Stamets identifies each new transfer from the previous culture with a "P Value" in which each new transfer gets the next higher number in the sequence of the culture. If I am wrong about this and do not understand the P value system please chime in on this thread and explain to me how the P value works.  I do not want to spread wrong information.  Until then I will say that the agar dish that I am working with in this grow was dish P3.  I allowed the P3 Golden Teacher LC to colonize.  Once the mycelium in he LC had established itself I gave it a good shake to break up the mycelium and help speed up the colonization time. On 10-11-13 I inoculated a total of four quart jars which were filled with wheat berries with about 2cc of LC each.  The jars were prepared just like RogerRabbit does in his Let's Grow Mushrooms video.

As of 11-29-13 only one of the jars had reached full colonization.  The other three were close behind but they still haven't reached full colonization.

In this grow I am working with a tub half the size of the standard monotub. It only needs a total of eight quarts of spawn and substrate to fill the tub up to the four inch mark.  I am not 100% sure which book it is in but I am pretty sure that in "The Mushroom Cultivator: A Practical Guide to Growing Mushrooms at Home" Paul Stamets say that 10 to 1 is a good working ratio for spawn to substrate.  With this in mind I decided that one quart of spawn would work just fine with seven quarts of substrate.

For this tub I went with the Damion's Elementary Coir Tek. Before I started the tek I turned my flowhood on in my grow room to help clean the air in my grow room.  I let the substrate sit in the bucket for an hour then I gave it a good shake to mix everything up.  After I shook the bucket I let it sit for another half hour.  After letting the substrate to do it's thing for an hour and a half I dumped the substrate in the tub in front of my flowhood.  I placed a meat thermometer in the tub to monitor the substrate temperature.  I left the tub to sit in front of the running flowhood until the temperature of the substrate had dropped down to 100F.

Once the temperature had dropped down to 100F.  I broke up the colonized wheat in the grain jar and poured it in to the tub.  I mixed it all up, put the lid on, labeled the tub with the strain, the date it was inoculated on, cracked a beer and lit a smoke.  My fingers are crossed that all goes well and in about a month or so I will have plenty of fatty Golden Teacher shrooms to print and add to my print collection.  Even if this tub ends up contaminated and doesn't make it to harvest I still have a clean agar culture to fall back on, and I still have three of the four LC jars colonizing.  I also have the jar that I inoculated with culture P4.  Either way I will get the prints I need:thumbup:

12-5-13:

The tub as of 12-17-13 in the morning

And here is some pin porn of the tub after I opened it on 12-17-13

How long did it take for it to colonize? The grain bags have been trying to colonize for 2 weeks and 2 days and no white has been seen in the grain bags. Does golden teacher take a while? What causes it to become contaminated?

I don't know why people use such big jars? Just use more little jars instead of a few big jars. It's easier to deal with and they colonize faster.

:lolwut:

He's using quart jars, the de facto standard for..just about everything aside from BRF cakes

Nice flush, WW!

I would agree with cronicr that the coir is fine in open air and there's no need to spawn in front of the flowhood.

Save your sterile technique for when it matters, like your grains and cultures.

I have 22 quarts colonizing at the moment and im not even really ramped up, I could not imagine trying to keep track of 88 half pints. Im sure serious guys like Frank would lose their minds trying to keep track of 4x the number of jars they have at any given moment.

:whathesaid:

Plus, each jar is an opportunity for a loss.  So I try to minimize the number of containers I need to use at a given time.

Everything I do starts with live culture on an agar wedge.

My master quart jars colonize in 2 weeks at 81F.

I can fit 56 of these jars in my 35 gallon tub I use for incubation.

Each master jar is G2G'd into a filter bag containing 5 more quarts of grain. This colonizes in 7-10 days, tops, at 72F.

I can fit six of these bags into a single 35 gallon tub for colonizing.

With these time frames, I go from culture to harvest in about 5-6 weeks, running 10-20 monos a month.

I think I'm pretty set with my "large" jars

Im about to inoculate 12 jars, and I rather have 12 small ones then a few big jars..Because it increases the chances of atlast one jar being contaminated free.

I sincerely hope you're not hoping to get at least one jar contam free. 12 small or 12 large your chances depend on your skill.

And that is sound logic as a beginner, but that is not what you said.

Plus it would never fly for a large-scale cultivator. That is why you practice sterile technique until you get your contam rate down, to minimize loss

Actually less jars that are bigger will lower your contam rate. It' statistics

Also grain jars aren't like brf jars. A quart grain jar will colonize just as fast as a pint because you can shake grains

The jar was inoculated via liquid culture on 10-11-13.  I didn't get but one fully colonized jar out of 4 total until 11-29-13.  So ya it did colonize slow, but it was colonizing in a place that was pretty cold compared to the typical 80f that cubes love.  That has been fixed by adding an oil heater to the grow room that is running 24/7 right now.  The heater is placed under the metal table that everything is colonizing on and the table heats to about 86f now.  Contaminates are caused from mold spores that are in the air failing on top of your live cultures, or your mycology tools when they are exposed to the open air.  It is also caused from mold spores that live through sterilization ie. you do not pressure cook your substrate long enough/at the proper pressure.  There are other ways to introduce contamination to your grows but this covers the majority of them.

I usually use manure that I pasteurize in quart jars.  I just wanted to go with the bucket tek to lessen tha chances of running into a contam.  I want to re up my GT print collection before I get too crazy with contam prone things.

Hey everyone this thread has kinda gone dead.  Here is my attempt to jump some life back into it.  I hadn't had the chance to get to my set up until today.  Once I got to see it I was surprised to see this.

The mushrooms are really small.  I am guessing it is do to genetics.  It's all good though because I am making 10 prints right now.  That was the entire purpose of this grow.  I needed to re-up my Golden Teacher prints and now I have 10 brand new ones to work with:thumbup:

And here's a little bit of pin porn to :whacker:

I am trying to get everything in place to do a bunch of agar work in the very near future.  I am sick of multi spore work. I want to get some big fat potent ass fruits for every cube variety that I am working with and then clone that shit.  I've got my flowhood set up and all.  I'm just floating back and forth between 3 different spots right now.  I noc one place.  Then it colonizes in another.  And then my tubs grow out in another.  It's a PITA but it's working.
