newbie here. I have some spore prints on foil.
Can I not use this tek to activate them? Like in a still box, scrape some spores into the agar and let the spores activate and check for contam?
After that, I was going to use blenderless liquid tek.

Flame sterilize an inoculation loop, cool it in your agar, wipe the loop onto print to pick spores up, swipe across agar in a z pattern.
Good luck!

You can also use a sterile swab the same way (except for the flame sterilizing ). You can PC q-tips after wrapping them in foil if you don't have sterile swabs.

If you want to weigh it, for this amount of agar you will want about 2-2.5 grams depending on how soft or firm you want it to be. If your using powdered agar and don't have a scale, a level tsp (5ml) is roughly 2.5 grams. Remember that no matter how much you want to mix up, you want a ratio of 20-25 grams of agar to 1 liter of water. If you have issues with the mix being too stiff while mixing then adjust the ratios for less agar. Some food grade agars will work out to be a little stiffer than others, some experimentation may be required.
two heaping tablespoons or teaspoons?

In the tek he uses solid agar so he meant tablespoons, but that's quite a bit for powder

That size looks good to us. PP5 plastic code, check. Just make sure the lid snaps on securely, cannot ascertain that feature from the photo. Happy Tissue work!

3.1 cm is very small for the diameter of the plate. I have 5.5 cm plates and I wouldn't go smaller.

Thank you very much for this tek. I managed to find some very cheap tubs, £1.50 for 8 . Seem quite sturdy too so should last a few runs . So the condensation on the side won't matter too much I wouldn't think?
Anyway great tek very easy . 👌

Thank you Pastywhyte for this monumental thread. I've been digesting it a little at a time for the past week and I'm on page 263 lol. I just can't believe how awesome the community is with several fine folks repeatedly answering the same questions to help new aspiring mycologists get started. You guys fucking rock.
I've got my first couple batches of pasty plates noc'd up, I'm not worried about condensation, I'm excited to be doing agar instead of the PF tek I did back in 2000, and I can't wait to enjoy the fruits of these labors!

Damn. It's been 10 years since this post gave me the confidence to do agar. This really standardized the no pour method.

Pastywhyte said: You could steam em but be aware that you will need much longer times. I would do at least 70 min and see how they do. Elevation will play a role. Finally you really should get a PC cause agar and grain are a nice combo.
They can absolutely be steamed. The above advice from the man himself is accurate though personally i would just do 90 minutes if you're going that far.
