so, once you've transferred a wedge into a new plate, do you toss the old agar? part of me wants to bury under some coir and poo and see what goes down.

i used to keep mine all the time, in case transfer went wrong or something. then ended up with fridge/tote full of used plates.
  if ur doing plate>qt>g2g u should save it tho, cause if its a great culture, u wanna be able to go back and use it again/slant it. thats for clones and isolates tho. if its just MS plates, then u can save or toss

word. it was ms plates. burried it in the shade with some wet hpoo and coir for funsies.
when the first transfer spreads, I look for sectors I like, transfer those again and then when they are fully colonized, just dump the whole damn thing in a master, yeah?

I picked up some agar agar from Whole Foods. It's in a powered form. Should I use less than the 2 tablespoons called for in the recipe. After adding 2 teaspoons of potato flakes and 1/2 cup water, the mixture is pretty thick. Any help would be appreciated.

The 2 tablespoon ratio was for the ground bars. If I use powedered agar I have to weigh it, I prefer about 2-2.5 grams for 120ml of water. 2 tablespoons of powdered agar would be far too much, probably closer to 1 is what you would want.

yeah, I went ahead and added another 1/2 cup of water and 2 teaspoons of potato flakes and the consistency looks better. I'm guessing this stuff shouldn't be runny, but more 'mashed potato' like?

After simmering it will be pretty smooth, like liquid jello. You don't want it too thick. It should probably be good now. If you have a scale and your using powdered agar, you want roughly 2% agar.

I use powdered agar from the health food store. I weigh mine out. 10 grams per 500ml of water, 7 grams of dextrose and 1 tablespoon of potato flake. I may start using more potato in the future so my cultures don't grow quite so fast. If I forget about a plate for a couple of days it gets completely over grown.

Plates came out looking good. Used 1 tablespoon of the powdered agar. I believe RR says in his videos that 1 tablespoon = 2 grams. My plan is to get good growth on them from a spore syringe I made last night and then use that to inoculate grain jars with. that way I hopefully won't waste any Rye berries to a contamed syringe.

u mean to just scrape the spores onto the plate? I thought about doing that, but read somewhere that using a syringe would be better. I should've just posted a question in the AMU thread beforehand

All powders have a different density. I usually use more spoonfuls of agar than I do elme for equal weights.
