It looks clean but very tomentose. I would probably transfer to see what might pop up but if its the only culture you have then make a master and transfer as well.

Inoculated 10 plates with B+ MSS and 6 with colonized B+ WBS from a fully colonized jar.
From the reactions I've garnered by using colonized WBS to inoculate media, I'm thinking I'll catch some flack.
A small hole was dug with a sterilized blade utensil into center of agar inoculated with MSS. Keeping the solution in the center was something I didn't give enough thought to before starting the process. That being said, I can't guarantee the solution didn't travel on the surface. I should've given this aspect more consideration!
How could this adversely effect the process? I've read that mycelium reaching the container wall should be avoided..

For me it just germinated all over the plate instead of from the center. Just transfer away to a fresh plate to make sure it's clean.

Colonized wbs is fine to expand on agar. Is a great way to really narrow genetics, especially if you're looking for isolates. Colonized grain to liquid media is what would catch you flack.

Having water all over the surface of your agar can make bacteria spread like wildfire. I wouldn't worry about it though, time and practice will solidify your technique.

My pleasure brother.
Was it that fast B+ master you had?

speaking of.
hey guys check this out. a 2008 sample of ape. this multi spore syringe was stored in the fridge until 2014. i then injected it into pf jars around february 2014. once colonized i kept 1 pf jar in the fridge from march 2014 until i slurried that jar again at the end of last month march 2015. so a full on 1 year of pf jar sitting in the fridge. i then slurried that pf jar ala liquids. once that jar colonized. i took one piece of wbs grain from the slurried jars with sterile tweezers and put it on agar. this is after 3 days. i transfered it tonight. i was only able to transfer one plate. ill do a few more next week when time permits.

It's the method of inoculation that I like the most when dealing with agar.

just did the math. holy shit, i had ordered 2 ape syringes. one i grew in this set of grows below in 2008. kept the other syringe for a rainy day 7 years later. strong growth for being that old, sitting in the fridge on a colonized pf jar. then slurried to bulk grain.

You could transfer from really anywhere on those plates. I would stick to the left side of that first plate though. The other side looks off to me.
IME Mexicana is slow as fuck on agar. It seems to perk up a bit on grain though.

Pasty, do you still wrap your entire rounds in alu foil or just a cap over the lid? Or anyone else?

I always just wrapped the lids. I don't bother with the paper towel either.

The foil goes around the sides alittle too. It's hard to sit them upright with foil going around the bottom IMO.

I wrapped the entire thing in foil, stacked 16 in a 16 qt PC and set a pot top on top of it all to keep things relatively even.
