People did this before pasty plates, this tek is kind of an upgrade.

So I got a question for you guys. I did this tek for the plates and used polyfil as I do not have any micropore tape. I used a syringe and and let the plate cool to room temp. then put my foil over the tops and pc'd them for 45@15psi. then let them cool to room temp today. I have condensation on the walls. do you guys keep the foil on until you use them or would it be ok to pull them off to help dry off the condensation? i think i read it will clear up in a few days. just didn't know how you guys handled it.

Condensation is ok, you let your pressure fluctuate too fast while cooking. Bring the pressure up slow and down slow, the problem will go away. I dunno what this talk of a syringe is about though.

I leave the foil on them until I have them in my SAB ready to use and don't even worry about the condensation anymore. I've tried every tip and trick suggested in this thread and never was able to get them to come out without condensation. It doesn't hurt anything. When you go to inoculate them remove the foil and over the course of the next week-ish (sometimes longer) the condensation will slowly go away. Some people like to wipe it off with the paper towel when they open the lid to inoculate, but I've found that to be an un-needed contam vector.
If you want to get a good look at what's going on in the plate while condensation is all over the walls preventing you from seeing in, hold the plate up near a ceiling light and look in through the bottom. I've spotted bacteria that way that I couldn't see from the sides/top even without condensation on the walls, so I personally think that's the best way to look at what's going on in them anyway.

I just wipe it off with the sterilized paper towel in my SAB when I go to use them. It also sounds like he inoculated and then PC'd the spores to death with the way he wrote it, heh.

I use blue food coloring. It's a fairly deep blue but I can still see through it well.
Actually I've got a pic that shows the color...
You can see it fairly well on the plates on the upper shelf.

This is bacteria, correct? I'm just wondering because there's no fucking way all 8 APE syringes have bacteria. I've had it before and I KNOW what bacteria looks like but I just need a second fucking opinion so I know I'm not losing my mind.

The other ones seem to have wet spots on them if you know what I mean? Water kind of slightly poking up above the agar.
Those are myc dots and a little of the water I was talking about:

This is the only APE I've got to germinate, should I transfer this:

Tissue sample from a cake on it's second transfer, look at that wicked bacteria:

Once again I reached my weekly upload limit so excuse the external links.

Yeah you should transfer that APE. Make a few transfers from it from on the left side of the 4 you've got written on it. Try taking tiny wedges and also try scraping some of the myc off the top to hopefully avoid the bacteria underneath. You should easily be able to get a clean culture out of that with just a transfer or two.

That's what I was thinking.
I just hope it's not a shitty culture. Really pissed I have 8 syringes and even some of the new ones are showing straight bacteria.

This is why you should always get syringes from our sponsors, or be very careful and make your own
If I'm trading with someone, I ask for a print. I've never gotten a syringe from another member that hasn't been gross with contams.
Obviously you can trust more experienced members but my golden rule for trading is prints for prints.

All bought from our trusted vendors ,
I don't trust anyone
EDIT:
I bought four originally from a sponsor and got nothing but bacteria on all plates except the APE 4.1 which is my second drop of APE from another syringe. Funny thing is the first drop contaminated. I contacted the vendor and said I couldn't identify anything but bacteria and they sent me four more.
Two of them are already showing bacteria. Guess I'll just clean this one up. Was hoping for some stronger growth though.

You should have told the sponsor that you saw bacteria under the scope and requested clean syringes. Our sponsors are usually great about that.
