Then you can't store it to a slant, and it can make for a crap shoot. Once you've got a proven isolate, it makes thing more consistent.

You could always cut out a section from the very center of your plate to make a transfer before blending up the rest of the plate as an LI. I did this once with some first transfers after I accidentally let them grow to the edge before being able to transfer out the different sectors. I basically just cut out the original donor wedge and put to a fresh plate to let it grow out again. If you don;t do slants, how do you plan to store cultures long term?

I used to just keep them on plates in my fridge. I watched the colour of the agar and when all The food coloring was gone I transferred to a new plate. Now I'm moving in to storing them as LCs and LIs made with distilled water.

Pasty,
I'm not 100% sure if I've got the figures right, do you know exactly how many ml the base recipe of this tek produces? I'm not sure that I have something to measure it to be sure, or if it's safe to just add up the sum of all the parts. Not sure how to convert the 2.5g of agar into a specific number of ml. I'm guessing it's less than 200 but I'm really not certain.

I do it kind of like this, probably a bad idea I know but it seems to work well, with Reishi I know for sure. Hell, I had a year old condiment cup I did this with, without it ever being in the fridge(long story, but it WAS poured deep, and I made one transfer, and it made it throught the first flush no problems at all!)
I just had a shitty harvest of a Golden Teacher clone, another long story that I don't feel like retyping, so if you care to read about it, you will have to go to the very last of my journal to see it, also there is a pic in there of the biggest tamp stone I have EVER harvested!
LOL, kind of seems like a ploy to get you to take a look at my journal, but it's not, I like to keep it small, but if anyone from here feels like taking a look and replying, I sure ould not mind, it is not the most exciting journal, and it is open to all, that is probabably why I don't get very many visitors, I can be rather boring and long winded, LOL again!

Alright I just ordered a big bag of MEA mix (couldn't find agar locally so figured I'd just get a big premade mix). Tomorrow I shall head to the store and get all my mini rounds and tape. Thanks for the tek pasty. Big bear is on board ready to fuck with some isolates!

Which should I isolate my KSSS strain from?


The top two are transfers and the bottom is a grain from my master jar.

Mini rounds are hard to find at grocery stores. I had to order mine off eBay. Agar, on the other hand, well, I found agar powder, dextrose powder, and potato flakes at vitamin cottage.

Nope, None have been fruited as of yet. I have a mono going with transfers from the Pasty plate and the one labeled KSSS grain colonizing and I just shot the LC into a BRF jar and it's colonizing well.
Ziploc Lock N' Twist work fine, just look for the PP5 label.

Around 2 months ago I dropped a big chunk of agar into LC water and let it hang out, swirling it around every few days. When it was broken up a bunch, I sucked it up into some syringes and put a drop or so on 3 agar plates to see if it was clean.
Here's what I got back:

So I'd say it's likely clean enough to use on grains since 2/3 came back pretty good? The other one I feel like I put too much in and it splattered because it's not smelling, it's all white, and the growth wasn't super quick like bac usually is. Is it possible it's clean, just tons of inoculation points due to it splashing around?

I have some plates like that, either as a result of spores being sloshed around or, on the odd occasion where I drop my transferred sample into the dish and it rolls around a bit before becoming stationary and growing into the plate.
Hard to say if that's clean or not without putting it to grain. Check it out and see.
