1. What's the specific reason for cutting out a piece to transfer if the whole thing looks good?
2. What do you mean by fastest growth? Just the biggest circle of mycelium there or a specific sector which looks to be pulling away quickest?

I mixed PE with PE 6, KSSS and Cambodian on 3 pasty plates. They all pined in just over a week.

Oooh I know this one. Smaller pieces reduce the chance of contaminates landing on your transferred bit. Also you're more likely to grab a specific chunk of genetic material if you make a small transfer from the edge where it's growing the fastest. This also helps ensure that what you just transferred is still in the vegetative colonization state, vs trying to fruit, where they kinda stop colonizing at that point, become harder to transfer. (not harder it just takes a day or so for them to switch modes, it seems like)

Thanks, man. Also my lids are purple and opaque, have paper towels stuffed on them, and tape covering the hole anyway.

I'm wary about removing the paper towel and infecting it since I don't have any micropore tape yet. Also the paper towels stick to the tape I use and cover the biggest portion of the lid. I'll try in my SAB next time though. You using a camera phone?
Here's a (rather large, sorry) GIF of me dropping an agar wedge into the distilled water thing for your LI Tek. I have a full video, but I think this is the most crucial part. Please critique!

By the way, an X-acto knife SUCKS for this. When you heat up the blade, it heats up the entire handle and is soo f'ing hot. Do scalpels do this?

That's the same exacto I use. It gets hot as fuck but I use that as motivation to work faster. You look very unsure of your movements but that will come with practice. Try practicing with empty plates and jars just to get your movements down. I made a video for you last knight but my computer was being gay and I couldn't get it off of my phone. I got pissed and deleted the video but I will make another one and try using my wife's computer to upload it.

Yeah, that was my first time cutting agar so I am still pretty new. My movements were kind of restricted because I was moving the X-acto around because it was burning the shit out of me, heh. Damn that hurt. I actually opted for a .GIF and not a video because I'm cussing in the video LOL.
I feel like my time between wedge in the air and into the jar was pretty solid though + I don't see my hands crossing either barrier too bad. I think if I contaminated anything, it wasn't during that part.
Can't wait for a video of yours!

I actually had two of them one of making an LI and the other was putting spores on agar and transferring petri pins to new plates. I shouldn't have deleted them but I was getting pretty pissed off at my computer. I'm kind of a luddite.

I'll get the wife to help me with it she's pretty good with computer shit. I will be making a few PE LIs this week as soon as my plates grow out a bit more. I wasn't planning on putting any spores on agar again but I have a few spare plates and lots of extra print so I can sacrifice them for the sake of a video.

I don't get to it every day. I'm pretty busy with work and kids but I like to spend at least a couple hours on it during the week and I generally spend my entire weekend on myco work. I would say its about 40 hours a week on average.

I'm just imagining your wife opening up the fridge and there's your bottles with mushrooms in them. She opens the microwave, more bottles. She pulls the blankets in your bed down to give you a kiss goodnight and there's 37 mushroom bottles in the outline of a male body instead.

That's not too far off. She likes it better than my old hobby (getting drunk) so the wife is pretty supportive when it comes to my mushrooms.
