I ended up letting some of my plates fully colonize and pin and I've just been transferring the biggest pin from each plate to new plates. Seems to be one way to isolate genetics easily 
The pins are so easy to stab with an x-acto and then just slide off inside the next plate.
I've finally gotten some of my agar onto grain and it's taking off without issue so far. A little slow maybe but I'm aware it has to learn to metabolize its new media.
edit: actually just checked and one of my second or third gen pin dishes shot out the biggest pin I've ever seen on agar.
I'm assuming if a pin can get big on agar it'll be massive on a proper bulk sub? Can't wait to transfer that sumbitch. It's sprouting up out of a dish I already transferred a wedge from.

I have done the same thing recently, but thinking it was in a sterile environment, I threw the whole mushroom in, not thinking about the cap containing spores, so my grow turned out to be ms anyway!

Pins don't typically have spores at their early stages.



The first two are this big ass pin I just mentioned and the third is one of the previous gen pins freshly transferred.
This is supposed to be APE, but some folks mentioned that most cubes can't pull their full color from the nutrients within agar so usually end up looking pale/albino. Can anybody confirm/attest? That pin looks PRETTY fuckin APE to me.

Also here's a contaminated plate that I took out of the glad container and took a picture of in my fingers.
 Failed transfer I guess. Is that bacteria?

You are absolutely correct, I had had these sitting up in a windowsill, and had not checked on them in at leat a month, so the pin I took was mature, and that def. Looks like bacteria in that second pic.
Good thing about that grow that turned out to be MS, is I took a very fast growing and good looking cluster, and cloned one of the fruits, I'm working on getting that to grain tonight.

I am sure you probably know this, but it seems to me you have some good mycellium on that cotamed plate, that if you were careful, and just did not get into the agar, and just got you a small sample of what appears to be good mycellium, you may be back in the game off that plate!

That would be false. I've got numerous nepal and pink buffalo plates that have been pinning, and they all look the same as any other nepal or pink buffalo pins with the proper cap color. What you have is a good looking in vitro pin that I would would pull and put to a fresh plate. It's a known fruiter now and shows the APE traits (that I guess) you're looking for.

Whoa I can't see myself waiting that long. I'll probably get that pin soon then. It's definitely got the traits I want.
Given that, what are the chances of it getting way bigger on a bulk sub?
Also @Iam fuck that contam plate, I have a couple good ones going right now so I have no reason to fuck with that.

Yea I'm not recommending to wait that long, I just have way to many plates sitting around and need to tighten up. If you do though, the spores it drops will also be sterile since they were in vitro as well, take as sterile swab to them, and you've got some clean spores.

Pins will have pigment and complete a life cycle invitro. That pin is albino

This seems like a really cool way to use agar to get spores. Honestly, agar gets neater and neater as the days go by.
