You cut wedges and transfer those. Loops are more for streaking spores. A good scalpel is essential. I use a metal exacto.

A wedge? What do you mean? Like a triangle of just myc or the agar it's growing on too?

Pudgy said: I did my first transfers today... I made my own scalpel which resulted in laughter and it was a total joke. Then I decided to use these hooks I use to detach exhaust pipe springs which worked okay.
I thought I was supposed to cut chunks of agar out and transfer them -- sigh... So one of my new agar plates has like an obliterated chunk of agar scattered across the top. One has an upside down triangle so the myc side of the triangle is on the surface of the new plate. The other two transfers were just surface mycelium with no agar chunks (proper).
After all of this, I really can see how an actual loop would be so nice to have.
I always take a piece of agar when making transfer. I cut a small wedge about the size of a grain of rice from the leading edge of growth. Loops are for putting spores on agar.
@ stareatclouds A coat hanger is a bit big but might work. I just stab my loop into the agar to cool the loop then I smear spores in and over the hole I made when cooling the loop.

I wish someone would post a really simple little video clip of these procedures.
I see, do you take the entire piece of agar (to the bottom of the plate) or just below where the myc is growing.

That depends on how thick the plate is. I just take as little mycelium and agar as I can get cleanly from the plate. RR's lets grow mushrooms videos have a section on this.

I have a real inoc. Loop that I got from O-G but have never got to use it, cause I misplaced it, like an idiot.

I watched those clips. I don't have $9 to spend on his videos, it's worth it to me and when I can I'll buy.
However, what you're saying makes perfect sense, I can just spoon a tiny layer into my minis.
On that note two of my four transfers are doing very well, I can see very tiny growth since yesterday.

I think the whole thing is on you tube for free and when you have the money buy it.

Wow, I'll look right now. Oh no doubt, I'm here for good -- this "hobby" has grabbed me hook line and sinker. It has little to do with tripping at this point. I'd love to own all of his work.

Hey hey hey, just found the answer to my condensation problem!!!!!
After the glads have cooled, if you have a lot of condensation shake and tap the glads till to collect most of the water together, turn the glads on thier lids then with the excess water swish away the condensation on the important bit so you can see. This is all done in the Sab befor Transfers so you can tip out excess b4 hand.

stareatclouds said: I was joking. I don't have a guitar. I know this is usually aimed at stopping something from growing, but is a metal coat hanger too big? Am I digging the loop into the agar or just pressing down into it?
Here's what I do. Just press it over the area you want transfer and twist. Obviously it's only for transferring small pieces but it's quicker and easier then trying to cut out and pick up a little chunk with a blade. The diameter of the loop should be about the same as the depth of the agar for it to work.

I'm just going to wipe it off with the sterilized paper towel that is stuffed on the lid. No more condensation.
Kizzle,
Thanks.

I did that as well, worked great -- even wiped the surface of the agar before noc. No tammies yet, growth looking good.

Kizzle,

Kizzle is blessed with great cleanliness. This is only for people who want to limit the exposure the SAB air who are beginners in agar and worried about their sterile technique.

I find if I leave my plates sitting at room temperature for a few days the condensation pools on the agar and I can just pour it onto the floor of my SAB at inoculation.

Thats what I said but use that to clear the lid area is really useful and limiting exposure.....

It seems as if you and I are bashing heads, if this is my fault, then I am sorry!
I have read every post in this thread, and I do remember you saying this was a great thread, for people to do, and then come back and report there findings for others to learn, I know you were just complimenting Kizzle, this is no place for petty bullshit, so again, if I offended you, I am sorry, and I respect the kind words you had for Kizzle.

I think you missed the bit I wrote above that about condensation and using it to clear your view. This Tek Rocks, you rock......we all rock....
I said something, Kizzle replied, we all moved on. Maybe my idea is a little stupid but it worked for me after I didnt wipe the lid.

Right on bro We are all noobs at this, once we quit being noobs, we stop learning! I do beleive that is an old RR quote, one of the very few times you will EVER hear me quote RR.
