you can find bars of agar agar in the Asian food section or you can get agar powder at health food stores.

Just thought I'd chime in.
Took clones a few days ago. Did the worst sterile technique ever. Felt clumsy as fuck. Just couldn't get the pieces to fall in correct.
No contamination yet and lots of solid growth. Still lovin' it Pasty. Keep it up.

If you live in the states, out-grow is a great source, so is amazon. I would go with the powder, but that is only because that is all I have ever worked with

Yup, I buy the agar-agar powder, then light malt powder, then add some nutritional brewers yeast(I think you are supposed to use a different type of yeast now, But the info in GG&MM said that's what to get, I just dry mix in a bowl, then add water, then personally I use v-tek screw tops, just loosen the lid a little, and tighten as u pull out of PC, no problems!

Pastywhyte said: Not sure about yer ratios but the standard 2% agar and nutes still apply. So for 500 ml water ya need 10 grams agar 10 grams nutes. I don't use yeast myself but something along the lines of 7grams potato flakes 2 grams karo and a gram yeast would give a reasonable mix. Less nutes for fast growth and more for storage or spore germination is the way I like to look at it. You can greatly exceed the 2% nutes and it still works, just might be a little slower/tomentose.
If I use dextrose instead of karo is that ok?

All my mazatapec plates are pinning in vitro. Can i take clones knowing they are clean from these plates? Or since its an isolate are they all gonna be the same anyways?

Invitro fruits are common with these, they will not be isolates as most clones are composite genetics, but they will be clean and are usually indicative of strong fruiting potential. Just transfer the pin and use that plate to inoculate and slant/store.
@Edmunter, dextrose will be fine instead of karo

Way cool to know. So what about the plate i transferred from? Does that mean the entire culture posesses fruiting potential as well or just the sector its fruiting on

My little mini rounds are still damp inside, the condensation isn't gone and I don't see any growth. It takes like 5 days in optimal conditions right?

Did you take the foil off?

I did take the foil off. The napkins were just damp, no actual wetness and the pore tape was dry.

Then they are probably good. Depending on the gel strength of your agar (it can vary with food grade somewhat) you might need a little more. Maybe a little less mix in each round might help as well. As for germination it can take anywhere from 3 days to never. 5 days might be an average but is by no means a standard. I have had spores simply refuse to germinate on many media types. Give it some time or inoculate more. When I get home I will link a great thread on pressure hydrating stubborn spores.

Okay.
I know the agar looks pretty damn stiff -- so maybe the agar powder was a little much or something.
I used potato flakes too so maybe that had something to do with it.
Live and learn, if it doesn't grow after 7 days I'll just start over!

I poured some really deep plates of agar back in Jan. , I had very small amounts of growth on a few plates, but got in some different prints and did not fuck with them again till about a month ago on one, and this was sitting at room temp. 2 of them that had small amounts of growth, I transferred to knew plates about months ago, and put them in the window, they were Gt and brazil, both fruited Invitro .
I have never had the problem of to stiff agar, but have had it not stiff enough, using the measurements from the v-tek, grain water agar, it made it really suck to try and transfer
