Going from spore to agar to grain without transferring seems a lot like going from spore straight to grain to me. The point of spore to agar is to clean the desirable mycelium through transfers. Do some transfers from the leading edge of your colonies and post your results is all the advice I can offer. I’m new to germ plates and just transferred from my first, I took 4 transfers from the best looking germ plate. Tried to select different looking mycelial growth, and the results will give me more insight into what was actually happening on the germ plate.

I also suggest diving into the well of knowledge provided by the fellow above, PastyWhyte. He is my agar guru <3

45 minutes is what I used to do before having a PC and I had no problems. With a PC and depending on the media used I sterilize from 20 to 30 minutes, mileage may vary depending on the altitude you live.

Give it one hour if you want, it won't damage anything and peace of mind is a good thing.

That's what I did in the beginning yes, but I cut the time down more and more over time until I realized 45 minutes were enough.

so im about to turn off my stove and start letting the PC cool. i saw that you typically allow it to cool overnight - how long should i wait before opening it up since its only 5pm right now? should i still wait till the morning to take them out or is a few hours fine?

I found a 12 pack of 1/2 pint Mason jars at smith's for only like $15 or less after tax.  That's not a bad price considering your buying glass jars that can be reused indefinitely.  Just thought I would mention it in for those wanting some kind of glass containers for their agar storage dishes

so when i woke up this morning and took out my plates i noticed i messed up and didnt have some of the containers flat enough. is the agar being tilted going to give me any serious issues or is it workaroundable?

It might be a pain if you’re dropping spore solution on it however it will still work. It won’t even make a slight difference for transfers or using a spore loop. I don’t worry about slanted agar myself.

Have you noticed a difference in the look of the myc on a slanted plate? When mine are slanted I seem to get more rhyzo on the thin side and more fluffy/cottony on the thick side. Does this sound reasonable?

Yes, I assume due to a lower concentration of nutes in the thin side. A small gradient likely forms.

Ok thanks... I re made some last night.

I took 3 tissue samples and used 2 plates to test two different liquid cultures i made..

Only thing I did different is I used these Tupperware containers instead, it was all I could find.



Should still work right?
